大口黑鲈弹状病毒弱毒冻干疫苗在溶解过程中存在活性衰减现象,为了保持疫苗在溶解过程中的生物活性,本研究通过筛选适宜的溶解剂以获得冻干疫苗的高效溶解活化剂.首先以PBS缓冲液、人参皂苷、β-葡聚糖、黄芪多糖、山莨菪碱等溶液溶解活化大口黑鲈弹状病毒弱毒冻干疫苗,并测定各组溶解活化后的病毒滴度(TCID50),初步筛选出溶解活化后滴度损失最小,病毒活性最高的活化剂;进一步通过比较不同类型人参皂苷溶解活化冻干疫苗的效果,确定最优的人参皂苷类型;最后用溶解活化后的各组疫苗分别免疫大口黑鲈,在鱼体内验证疫苗的保护效力,并测定免疫相关因子、干扰素-γ(IFN-γ)与白细胞介素-12(IL-12)的表达水平.研究结果显示,人参皂苷 Rg1活化冻干疫苗后病毒滴度最高,有效保持冻干疫苗的病毒活力;以 20 mg/mL 人参皂苷 Rg1溶解活化冻干疫苗并浸泡免疫大口黑鲈(Micropterus salmoides),相对免疫保护率高达 97.6%.同时,人参皂苷 Rg1溶解活化冻干疫苗组显著上调了脾脏、头肾和鳃组织中免疫相关因子 IFN-γ 和 IL-12的表达水平,其中脾脏组织中12 h达到峰值(IFN-γ 和IL-12分别上调至20.05倍、230.64倍),头肾组织中48 h达峰值(IFN-γ 上调 21.22倍),鳃组织中 4 h达峰值(IL-12上调至 384.87倍).综上所述,以 20 mg/mL人参皂苷 Rg1溶解活化的大口黑鲈弹状病毒(MSRV)0509弱毒冻干疫苗浸泡免疫效果佳,具备良好的应用前景.

This study aimed to identify an optimum diluent to mitigate activity loss during dilution of an attenuated freeze-dried vaccine against Micropterus salmoides rhabdovirus and to enhance its immunogenicity.The study initially evaluated PBS buffer,ginsenosides,β-glucan,astragalus polysaccharide,and scopolamine solutions for diluting and activation of the freeze-dried attenuated virus vaccine.Viral titers(TCID50)were assessed after activation for each group to identify the activator associated with the least titer reduction and the maximum viral activity.Subsequently,the optimal ginsenoside type was identified by evaluating the effects of different ginsenosides on the dilution and activation of the freeze-dried vaccine.Ultimately,each group receiving diluted and activated vaccine formulations was administered to Micropterus salmoides to assess in vivo vaccine efficacy and quantify immune-related parameters,including expression levels of interferon-γ(IFN-γ)and interleukin-12(IL-12).The results indicated that ginsenoside Rg1-activated lyophilized vaccines exhibited the highest viral titers,effectively maintaining viral activity.When the vaccine was diluted with 20 mg/mL ginsenoside Rg1 and administered by immersion,the relative immune protection rate in bass reached 97.6%.The ginsenoside Rg1-diluted and activated freeze-dried vaccination group significantly increased the expression levels of immune-related cytokines IFN-γ and IL-12 in the spleen,head kidney,and gill tissues.Expression peaked at 12 h in spleen tissue(IFN-γ and IL-12 increased by 20.05-fold and 230.64-fold,respectively),at 48 h in head kidney tissue(IFN-γ increased by 21.22-fold),and at 4 h in gill tissue(IL-12 increased by 384.87-fold).In summary,the use of 20 mg/mL ginsenoside Rg1 as the dilution activator for the MSRV 0509 live attenuated freeze-dried vaccine markedly enhances immunogenicity following immersion immunization,indicating strong application potential.
