乳源嗜冷菌及其耐热蛋白酶对乳制品品质的影响OA
The Effects of Psychrotrophic Bacteria in Milk and Their Heat-resistant Proteases on the Quality of Dairy Product
嗜冷菌可在0~5℃低温条件下生长繁殖,并会大量分泌胞外耐热蛋白酶,是造成原料奶及其制品腐败变质的主要诱因.耐热蛋白酶经乳制品常规热处理工艺加工后仍有部分活性残留,可持续分解乳中酪蛋白并与脂肪酶产生协同作用,导致乳制品出现苦味、凝块、分层等品质劣变问题,缩短产品货架期.本文系统梳理了乳源嗜冷菌的定义、菌群分类及污染途径,阐述了蛋白酶的分类、耐热特性及其对乳制品品质的作用机制,同时综述现阶段耐热蛋白酶主流检测方法(福林酚法、ELISA法)与嗜冷菌定量定性检测技术(传统平板培养法、PCR、流式细胞术、第三代基因测序技术)的应用现状,并对比各类方法的技术优势与局限性.此外,本研究结合最新技术进展,从源头防控、精准检测、残留耐热蛋白酶活性抑制三大维度展望未来研究重点,以期为乳制品行业实现嗜冷菌与耐热蛋白酶的高效精准管控、产品品质升级及货架期稳定保障,提供理论依据与实践技术支撑.
Psychrophilic bacteria can grow and reproduce at low temperatures ranging from 0 to 5℃and secrete large amounts of extracellular heat-resistant proteases,making them a major cause of spoilage and deterioration of raw milk and its products.Even after conventional heat treatment processes used in dairy production,some residual activity of these heat-resistant proteases remains.These residual proteases continuously break down casein in milk and act synergistically with lipases,leading to quality issues such as bitterness,coagulation,and stratification in dairy products,thereby shortening their shelf life.This paper systematically reviewed the definition,bacterial classification,and contamination pathways of lactobacillus-derived psychrophilic bacteria,and elaborates on the classification,thermostable characteristics,and mechanism of action of proteases on dairy product quality.At the same time,it summarized the current application status of mainstream detection methods for thermostable proteases(Folin-Phenol method,ELISA method)and quantitative and qualitative detection techniques for psychrophilic bacteria(traditional plate culture method,PCR,low cytometry,third-generation gene sequencing technology),and compares the technical advantages and limitations of each method.In addition,this study combined the latest technological progress and from three dimensions of source control,precise detection,and inhibition of residual heat-resistant protease activity,looks forward to future research priorities,in order to provide theoretical basis and practical technical support for the dairy industry to achieve efficient and precise control of psychrophilic bacteria and thermostable proteases,product quality upgrade,and stable shelf life guarantee.
