Genetic characterization of H9N2 avian influenza virus from live poultry markets in Changsha,2022-2023

目的 分析2022-2023年长沙市活禽市场(Live poultry market,LPM)H9N2亚型禽流感病毒(Avian influenza vi-rus,AIV)血凝素(Hemagglutinin,HA)和神经氨酸酶(Neuraminidase,NA)的遗传进化特征及关键分子变异,为防控H9N2 亚型AIV疫情提供实验室数据.方法 采集长沙市活禽市场的环境及禽类样本809份,采用实时荧光RT-PCR方法检测甲型流感病毒及H9亚型核酸,对H9阳性样本进行病毒基因全长扩增和测序,利用生物信息学软件进行序列比对和遗传特征分析.结果 核酸检测结果显示环境中AIV的阳性率为64.15%,其中H9总体阳性率最高为48.08%,混合感染中H5+H9的总体阳性率最高为5.07%.遗传进化分析显示,HA基因进化分析表明,29株样本属于B4.7.2分支,5株属于B4.7.3分支.NA基因均属于欧亚分支的Y280-like亚群,与候选疫苗株A/Anhui-Lujiang/39/2018亲缘关系较近.HA蛋白裂解位点为PSRSSRGLF,符合低致病性禽流感病毒(Low pathogenic avian influenza virus,LPAIV)特征,但存在Q226L(100%)、I155T/N(17.65%/82.35%)及G192R突变(100%),提示潜在的哺乳动物适应性.NA蛋白茎部63~65位点缺失,关键耐药位点未发生变异.糖基化位点分析显示,毒株具有6个保守位点(29、141、298、305、313、492),与部分参考株相比,缺失105、145、206、218位点,且新增313位点.抗原位点 分 析 发 现,61.11%(11/18)的HA抗原位点发生变异,可能导致抗原性改变.结论 长沙市活禽市场的H9N2 AIV虽为LPAIV,但存在哺乳动物适应性变异和抗原漂移,需持续监测其公共卫生风险.

This study analyzed the genetic characteristics of the hemagglutinin(HA)and neuraminidase(NA)genes of H9N2 subtype avian influenza virus(AIV)in live poultry markets(LPMs)in Changsha City during 2022-2023,to provide laboratory data to facilitate control of H9N2 subtype AIV outbreaks.A total of 809 environmental and poultry samples were collected from Changsha LPMs.Real-time fluorescence RT-PCR was used to detect influenza A virus and H9 subtype nucleic acids.Full-length gene amplifi-cation and sequencing were performed on H9-positive samples.Bioinformatics software was used for sequence alignment and genetic feature analysis.Nucleic acid detection revealed a 64.15%positivity rate for environmental AIV,and H9 showed the highest overall positivity rate(48.08%).Mixed infections exhibited the highest overall positivity rate of 5.07%for H5+H9.Genetic evolution analysis revealed that 29 isolates belonged to the B4.7.2 lineage,and five belonged to the B4.7.3 lineage,according to HA gene analysis.All NA genes belonged to the Y280-like subclade of the Eurasian lineage and showed close phylogenetic relationships with the candidate vaccine strain A/Anhui-Lujiang/39/2018.The HA protein cleavage site was PSRSSRGLF,in agreement with low pathogenic avian in-fluenza virus(LPAIV)characteristics.However,the presence of mutations Q226L(100%),I155T/N(17.65%/82.35%),and G192R(100%)suggested potential mammalian adaptation.The NA protein stem region exhibited a deletion at positions 63-65,but no muta-tions were observed at key drug resistance sites.Glycosylation site analysis revealed six conserved sites(29,141,298,305,313,and 492),whereas sites 105,145,206,and 218 were absent with respect to some reference strains,and site 313 was newly added.Antigenic site analysis revealed that 61.11%(11/18)of HA antigenic sites underwent mutations potentially leading to antigenic changes.Although H9N2 AIV from Changsha LPMs was classified as LPAIV,it exhibited mammalian adaptation mutations and anti-genic drift.Therefore,ongoing monitoring of its public health risks is necessary.
