目的 了解福建省部分地区猪肉源金黄色葡萄球菌(简称金葡菌)的流行特征、耐药性及耐药基因携带情况,为猪肉源金葡菌耐药性风险评估提供参考依据.方法 2023-2024年从福建省部分地区的农贸市场、超市和屠宰场采集猪肉样品,使用选择性培养基BP琼脂分离金黄色葡萄球菌,并进行PCR鉴定.采用琼脂稀释法测定其对21种抗菌药物的敏感性;并利用全基因组测序技术对其进行耐药基因及多位点序列分型(MLST)检测.结果 529份样品中共检出113株金黄色葡萄球菌,总分离率为21.4%.药敏试验结果显示多重耐药菌占比为42.5%,其中对四环素、磺胺甲噁唑/甲氧苄啶、阿奇霉素、青霉素和克林霉素具有较高的耐药率,耐药率分别为45.1%、45.1%、46.9%、50.4%和57.5%.对分离株进行基因组测序,共检出大环内酯类、氨基糖苷类、四环素类等9大类35种耐药基因,且所有的分离株均携带耐药基因,其中较特殊的mecA、cfr等耐药基因也被检出,同时还发现有34.5%的分离菌株携带12种耐药基因,占比最高.MLST分型分析发现,共存在15种ST型,其中ST7为优势ST型(34.5%).结论 福建省部分地区的猪肉存在严重的金黄色葡萄球菌污染,对临床常见药物表现出较高的耐药率,并携带大量的耐药基因,应加强对本地区猪肉源金黄色葡萄球菌的持续监测.

This study investigated the epidemiological characteristics,drug resistance,and drug resistance gene carriage of Staphylococcus aureus from pork in select areas of Fujian Province,to provide a reference for drug resistance risk assessment of Staphy-lococcus aureus from pork.Pork samples were collected from farmers´ markets,supermarkets,and slaughterhouses in select areas of Fujian Province from 2023 to 2024.Staphylococcus aureus was isolated with BP agar selective medium and identified by PCR.The sen-sitivity of the agar dilution method to 21 antimicrobial drugs was determined.Whole genome sequencing technology was used to detect drug resistance genes and perform multi-locus sequence typing(MLST).A total of 113 strains of Staphylococcus aureus were de-tected in 529 samples,and the total isolation rate was 21.4%.Antimicrobial susceptibility testing indicated that 64.6%of the bacte-ria were multidrug-resistant,and high resistance rates to tetracycline,sulfamethoxazole/trimethoprim,azithromycin,penicillin,and clindamycin(45.1%,45.1%,46.9%,50.4%and 57.5%,respectively)were observed.Genome sequencing of the isolates de-tected 35 drug resistance genes in nine categories,including macrolides,aminoglycosides,and tetracyclines.All isolates carried drug resistance genes,including special drug resistance genes such as mecA and cfr,notably,34.5%of the isolates carried 12 drug resistance genes.MLST typing analysis indicated 15 ST types,among which ST 7 was dominant(34.5%).Pork in the investigated areas of Fujian Province showed substantial contamination with Staphylococcus aureus that had high resistance rates to common clini-cal drugs and carried many drug resistance genes.Therefore,continued monitoring of Staphylococcus aureus from pork in this region should be strengthened.
