目的 采用不同培养方法制备2种卡介苗(BCG)冻干疫苗,比较2种疫苗的免疫原性.方法 BCG菌株分别接种苏通培养基静置培养和7H9完全培养基振荡培养,冷冻干燥制成BCG菌膜疫苗(BCG-BV)和BCG菌液疫苗(BCG-BLV).两种疫苗分别皮下免疫BALB/c小鼠,ELISA法检测血清抗体水平;流式细胞术检测小鼠肺部T细胞亚群;ELISA法检测脾细胞体外刺激后分泌细胞因子和炎症因子水平.结果 相同重量下BCG-BLV中活菌数比BCG-BV多.BCG-BV诱导血清抗BCG抗原和抗Ag85B抗原抗体水平升高;BCG-BLV诱导抗Ag85B抗体水平有升高趋势,亚类水平升高.BCG-BV可诱导肺部CD8+T细胞百分比显著升高,而BCG-BLV则降低CD8+T细胞百分比.BCG-BV和BCG-BLV组均可以诱导Th1(IFN-γ、IL-2)和Th2(IL-10)型细胞因子分泌增加,且BCG-BLV免疫组的水平更高.BCG-BV和BCG-BLV均可以诱导IL-1β、TNF-α和IL-6等训练免疫特征性细胞因子的分泌增加,且BCG-BLV组诱导IL-1β分泌水平更高.结论 不同制备工艺影响BCG冻干疫苗的免疫原性,丰富培养基且振荡培养的BCG-BLV冻干疫苗免疫原性更强.

Two types of BCG freeze-dried vaccines were prepared using different cultivation methods,and the immunogenicity of the two vaccines was compared.BCG strain was inoculated into Sutton medium and 7H9 complete medium respectively.BCG bacterial film vaccine(BCG-BV)and BCG bacterial liquid vaccine(BCG-BLV)were prepared by freeze-drying.BALB/c mice were immu-nized subcutaneously with the two vaccines,and the sera antibody levels were detected by ELISA.The lung T cell subsets of mice were detected by flow cytometry.The cytokines and inflammatory factors secreted by splenocytes stimulated in vitro were detected by ELISA.BCG-BLV contained a higher number of viable bacteria than BCG-BV.BCG-BLV induced an increase in the antibody level against Ag85B,and the subclass levels also showed a significant increase.BCG-BV induced a significant increase in the percentage of CD8+T cells in lung,whereas BCG-BLV decreased it.Both BCG-BV and BCG-BLV groups could induce the secretion of Th1(IFN-γ,IL-2)and Th2(IL-10)cytokines,and the levels of BCG-BLV immunized group were higher.Both BCG-BV and BCG-BLV could in-duce IL-1β,TNF-α and IL-6,which were characteristic cytokines of training immunity,and the BCG-BLV group induced a signifi-cantly higher level of IL-1β release.Different cultivation methods significantly influenced the immunogenicity of BCG.The freeze-dried BCG-BLV vaccine prepared in the enriched medium exhibited stronger immunogenicity.
