目的 分离并鉴定上海市金山区三带喙库蚊样本中的坦布苏病毒,并系统分析现有坦布苏病毒的基因型特征、遗传多样性、宿主适应性及分子流行特征,为病毒监测与预警提供科学依据.方法 2024年8月7-8日在某牛场采集蚊虫样本,经RT-qPCR检测、细胞培养、PCR检测、Sanger测序和宏基因组测序鉴定病毒株,获得全基因组序列.结合NCBI数据库中177条参考序列,提取ORF区域,采用系统发育分析、核苷酸与氨基酸相似性分析、滑动窗口分析及氨基酸突变谱分析,评估不同基因型间的遗传差异与生物学特征.结果 从三带喙库蚊样本中分离获得1株坦布苏病毒,属于Cluster 3.2型.系统发育分析显示坦布苏病毒可划分为Old lineage、Cluster 1、Cluster 2.1、Cluster 2.2、Cluster 3.1和Cluster 3.2六个基因型.不同基因型在宿主适应性、地理分布和分子特征上存在显著差异,其中Cluster 3.2具有最高宿主多样性.各基因型存在大量特异性氨基酸突变,尤其在E蛋白中发现了潜在关键位点(E-150N、E-365P、E-369G),这些突变可能与病毒生物学特性变化相关.结论 坦布苏病毒呈现出明显的基因型分化、宿主扩展及地域分布特征,特定氨基酸突变可能驱动其表型多样性及跨物种感染能力.本研究为坦布苏病毒的遗传进化分析及防控策略制定提供了重要依据.

This study was aimed at isolating and identifying Tembusu virus(TMUV)from mosquito samples collected a cattle farm in the Shanghai region on August 7 and 8,2024,and systematically analyzing their genotypic characteristics,genetic diversity,host adaptability,and molecular epidemiological features,to provide a scientific basis for viral surveillance and early warning.Culex tritaeniorhynchus mosquito samples were collected and subjected to RT-qPCR detection,cell culture,PCR amplification,Sanger se-quencing,and metagenomic sequencing to identify TMUV strains and obtain full genome sequences.A total of 177 reference se-quences from the NCBI database were included for comparative analysis.Open reading frame(ORF)regions were extracted.Phyloge-netic analysis,nucleotide and amino acid similarity analysis,sliding window analysis,and amino acid mutation profiling were per-formed to evaluate genetic divergence and biological characteristics among genotypes.The isolated TMUV strain from C.tritaeniorhyn-chus mosquitoes belonged to the cluster 3.2 genotype.Phylogenetic analysis revealed that TMUV could be divided into six genotypes:old lineage,cluster 1,cluster 2.1,cluster 2.2,cluster 3.1,and cluster 3.2.Significant differences in host adaptability,geographic distribution,and molecular characteristics were observed among genotypes.Cluster 3.2 exhibited the highest host diversity.Numerous genotype-specific amino acid mutations were identified,particularly in the E(envelope)protein,in which potential key sites such as E-150N,E-365P,and E-369G may be associated with changes in viral biological properties.TMUV was found to exhibit genotypic differentiation,host expansion,and regional specificity.Specific amino acid mutations might drive phenotypic diversity and interspe-cies transmission potential.This study provides critical insights into the genetic evolution of TMUV,and offers valuable guidance for surveillance and control strategies.
