2023年山东省蚊虫中部分蚊媒病毒携带情况分析OA

目的 了解山东省蚊媒病毒感染状况和分布规律,为当地蚊媒病毒的监测提供科学依据.方法 2023年8月在山东省6个监测点使用手持吸蚊器或紫外诱蚊灯两种方式捕捉蚊虫,将其分类计数后,每25~50只左右蚊虫作为一份样本进行分管研磨,提取总RNA,使用荧光定量PCR进行8种蚊媒病毒核酸检测,包括库蚊黄病毒(Culex flavivirus,CxFV)、Kadipiro病毒(Kadipiro virus,KDV)、版纳病毒(Bannavirus,BAV)、盖塔病毒(Getah virus,GETV)、Tahyna病毒(Tahyna virus,TAHV)、辽宁病毒(Liao ning virus,LNV)、西尼罗病毒(West Nile virus,WNV)和登革病毒(Dengue virus,DENV).结果 研究期间采集6种蚊虫14 813只.其中骚扰阿蚊6 721只,(45.37%),三带喙库蚊5 966只(40.28%),淡色库蚊1 851只(12.50%)为主要优势蚊种.将所有蚊虫分为360份样本进行检测,共检出Kadipiro病毒核酸阳性蚊虫样本40份,其中16份来自三带喙库蚊,18份来自骚扰阿蚊,3份来自淡色库蚊,2份来自中华按蚊,1份来自致倦库蚊;检出库蚊黄病毒阳性样本8份,其中三带喙库蚊和淡色库蚊各3份,骚扰阿蚊2份.2023年蚊虫Kadipiro病毒的最小感染率为2.70‰,Ct值为24.57~37.03;库蚊黄病毒最小感染率为0.54‰,Ct值为16.90~35.00.结论 山东省蚊虫中Kadipiro病毒和库蚊黄病毒存在较高的携带率,应进一步加强蚊虫所携带病毒的调查研究工作,为蚊媒病毒的防控提供数据支撑.

To understand mosquito-borne arbovirus infections and distributions in mosquitoes,we conducted mosquito surveil-lance at 6 sentinel areas in Shandong Province in August 2023.Mosquitoes were collected with handheld aspirators or ultraviolet light traps,morphologically identified,and counted.Approximately 25-50 mosquitoes were pooled per sample,homogenized,and sub-jected to total RNA extraction.RNA of the following eight mosquito-borne viruses was detected with real-time PCR:Culex flavivirus(CxFV),Kadipiro virus(KDV),Bannavirus(BAV),Getah virus(GETV),Tahyna virus(TAHV),Liao ning virus(LNV),West Nile virus(WNV),and Dengue virus(DENV).A total of 14 813 mosquitoes were collected and were identified as 6 species,among which Armigeres obturbans(6 721,45.37%),Culex tritaeniorhynchus(5 966,40.28%),and Culex pipiens pallens(1 851,12.50%)were the most common.Of the 360 pools of mosquitoes,40 pools were positive for KDV RNA:16 pools of Culex tritaeniorhynchus,18 pools of Armigeres obturbans,3 pools of Culex pipiens pallens,2 pools of Anopheles sinensis,and 1 pool of Culex quinquefasciatus.Eight pools were positive for CxFV RNA:3 pools of Culex tritaeniorhynchus,3 pools of Culex pipiens pallens,and 2 pools of Armigeres obturbans.The overall prevalence of KDV infection was 2.70 per 1 000 mosquitoes(Ct:24.57-37.03),and the CxFV infection rate was 0.54 per 1 000 mosquitoes(Ct:16.90-35.00).Mosquitoes in Shandong Province carried KDV and CxFV.Enhanced surveillance of KDV and CxFV will be essential to support the prevention and control of mosquito-borne viruses.
