白藜芦醇通过RhoA/ROCK通路抑制缺血性脑卒中大鼠神经炎症OA
Effects of resveratrol on neuroinflammation in ischemic stroke rats by regulating RhoA/ROCK signaling pathway
目的:探讨白藜芦醇调节Ras同源基因家族成员A(RhoA)/Rho激酶(ROCK)通路对缺血性脑卒中(IS)大鼠认知障碍和神经炎症的影响.方法:成功造模的72只大鼠按随机数字表法分为IS组、白藜芦醇组、RhoA激活剂(溶血磷脂酸)组、白藜芦醇+溶血磷脂酸组,每组18只.另取18只正常大鼠作为对照组.检测大鼠神经功能缺损评分、通过平台数、逃避潜伏期、脑梗死体积占比变化;免疫荧光染色检测海马CA1区脑组织离子钙结合衔接分子1(Iba1)阳性细胞数;ELISA检测海马CA1区脑组织TNF-α、单核细胞趋化因子-1(MCP-1)、IL-6水平;Western blot检测海马CA1区脑组织RhoA、ROCK1、ROCK2蛋白.结果:与对照组相比,IS组通过平台数减少,逃避潜伏期延长,神经功能缺损评分、脑梗死体积占比、海马CA1区脑组织Iba1阳性细胞数、TNF-α、MCP-1、IL-6及RhoA、ROCK1、ROCK2蛋白水平升高(P<0.05);与IS组相比,白藜芦醇组大鼠通过平台数增多,逃避潜伏期缩短,神经功能缺损评分、脑梗死体积占比、海马CA1区脑组织中Iba1阳性细胞数、TNF-α、MCP-1、IL-6及RhoA、ROCK1、ROCK2蛋白水平降低,溶血磷脂酸组对应指标变化趋势与白藜芦醇组相反(P<0.05);与白藜芦醇组相比,白藜芦醇+溶血磷脂酸组大鼠通过平台数减少,逃避潜伏期延长,神经功能缺损评分、脑梗死体积占比、海马CA1区脑组织Iba1阳性细胞数、TNF-α、MCP-1、IL-6及RhoA、ROCK1、ROCK2蛋白水平升高(P<0.05).结论:白藜芦醇可能通过抑制RhoA/ROCK通路抑制IS大鼠小胶质细胞活化及神经炎症,改善认知障碍.
Objective:To investigate effects of resveratrol on cognitive impairment and neuroinflammation in ischemic stroke(IS)rats by regulating Ras homolog gene family member A(RhoA)/Rho kinase(ROCK)pathway.Methods:A total of 72 rats successfully modeled were divided into IS group,resveratrol group,RhoA activator(lysophosphatidic acid)group and resveratrol+lysophosphatidic acid group according to random number table method,with 18 rats in each group.Another 18 normal rats were selected as control group.Changes in neurological deficit score,number of platforms passed through,escape latency,and proportion of cere-bral infarction volume of rats were detected.Immunofluorescence staining was applied to detect number of ionized calcium-binding adapter molecule 1(Iba1)positive cells in brain tissue of hippocampal CA1 region.ELISA was applied to detect levels of TNF-α,monocyte chemoattractant protein-1(MCP-1)and IL-6 in brain tissue of hippocampal CA1 region.Western blot was applied to detect RhoA,ROCK1 and ROCK2 proteins in brain tissue of hippocampal CA1 region.Results:Compared with control group,number of platform crossing was decreased,escape latency prolonged,neurological dysfunction score,proportion of cerebral infarction volume,number of Iba1 positive cells in hippocampal CA1 brain tissue,levels of TNF-α,MCP-1,IL-6,RhoA,ROCK1 and ROCK2 proteins were increased in IS group(P<0.05).Compared with IS group,number of platform crossing was increased,escape latency shortened,neurological dysfunction score,proportion of cerebral infarction volume,number of Iba1 positive cells in hippocampal CA1 brain tissue,levels of TNF-α,MCP-1,IL-6,RhoA,ROCK1 and ROCK2 proteins were decreased in resveratrol group,trend of changes in corresponding indicators in lysophosphatidic acid group was opposite to resveratrol group(P<0.05).Compared with resveratrol group,number of platform crossing was decreased,escape latency prolonged,neurological dysfunction score,proportion of cerebral infarc-tion volume,number of Iba1 positive cells in hippocampal CA1 brain tissue,levels of TNF-α,MCP-1,IL-6,RhoA,ROCK1 and ROCK2 proteins were increased in resveratrol+lysophosphatidic acid group(P<0.05).Conclusion:Resveratrol may inhibit activation of microglia and neuroinflammation in IS rats by inhibiting RhoA/ROCK pathway,improving cognitive impairment.
