目的:探讨山奈酚调控TLR4/NF-κB通路对类风湿关节炎(RA)大鼠炎症损伤的影响.方法:注射牛Ⅱ型胶原乙酸与等量弗氏完全佐剂、弗氏不完全佐剂混合液制备RA大鼠模型,制备成功大鼠随机分为RA组、山奈酚低、高剂量组、甲氨蝶呤组、高山奈酚+激活剂组,注射等体积生理盐水的10只大鼠为对照组,干预结束后,观察大鼠四肢关节肿胀程度并进行关节炎指数评分,并测量踝关节直径;ELISA检测IL-6、IL-1β、TNF-α水平;分离脾脏和胸腺,计算免疫器官指数;收集踝关节滑膜组织,HE染色检测滑膜组织病理学变化,Western blot检测滑膜组织TLR4/NF-κB通路相关蛋白表达.结果:RA组大鼠关节炎指数、踝关节直径、血清IL-6、IL-1β、TNF-α水平、脾脏指数、胸腺指数、滑膜组织TLR4、p-NF-κB p65/NF-κB p65表达较对照组显著升高(P<0.05);山奈酚低、高剂量组、甲氨蝶呤组大鼠关节炎指数、踝关节直径、血清IL-6、IL-1β、TNF-α水平、脾脏指数、胸腺指数、滑膜组织TLR4、p-NF-κB p65/NF-κB p65表达较RA组显著降低,不同山奈酚组间有差异(P<0.05);高山奈酚+激活剂组大鼠关节炎指数、踝关节直径、血清IL-6、IL-1β、TNF-α水平、脾脏指数、胸腺指数、滑膜组织TLR4、p-NF-κB p65/NF-κB p65表达较山奈酚高剂量组升高(P<0.05).结论:山奈酚通过抑制TLR4/NF-κB通路缓解RA大鼠炎症损伤.

Objective:To investigate effect of kaempferol on inflammatory injury in rheumatoid arthritis(RA)rats by regulating TLR4/NF-κB pathway.Methods:RA rats were prepared by injecting a mixture of bovine type Ⅱ collagen acetic acid and equal amounts of Freund´s complete adjuvant or Freund´s incomplete adjuvant.Successfully prepared rats were randomly assigned into RA group,low and high doses kaempferol group,methotrexate group,and high kaempferol+activator group.Ten rats injected with same volume of physiological saline were used as control group.After intervention,degree of joint swelling in limbs of rats was evaluated,arthritis index score was performed,and ankle joint diameter was measured.ELISA was applied to detect levels of IL-6,IL-1β and TNF-α.Spleen and thymus were separated to calculate immune organ indexes.Synovial tissue of ankle joint was collected.HE staining was applied to detect pathological changes in synovial tissue,Western blot was applied to detect TLR4/NF-κB pathway related protein expression in synovial tissue.Results:Arthritis index,ankle joint diameter,serum IL-6,IL-1β,TNF-α levels,spleen index,thymus index,TLR4,p-NF-κB p65/NF-κB p65 expressions in synovial tissue of rats in RA group were obviously higher than control group(P<0.05).Arthritis index,ankle joint diameter,serum IL-6,IL-1β,TNF-α levels,spleen index,thymus index,TLR4,p-NF-κB p65/NF-κB p65 expressions in synovial tissue of rats in low and high doses kaempferol groups,and methotrexate group were obviously lower than RA group,and there were differences among different kaempferol groups(P<0.05).Arthritis index,ankle joint diameter,serum IL-6,IL-1β,TNF-α levels,spleen index,thymus index,TLR4,p-NF-κB p65/NF-κB p65 expressions in synovial tissue in high kaempferol+activator group were higher than high kaempferol group(P<0.05).Conclusion:Kaempferol alleviates inflammatory injury in RA rats by inhibiting TLR4/NF-κB pathway.
