NDC80对宫颈癌细胞增殖、迁移和侵袭的影响机制研究OA
Study on the mechanism of NDC80 in regulating proliferation,migration,and invasion of cervical cancer cells
目的 探讨NDC80在宫颈癌中的表达及其对宫颈癌细胞增殖、迁移和侵袭的影响机制.方法 GEPIA数据库分析NDC80在宫颈癌及多种肿瘤中的表达.收集山西医科大学第三医院80例宫颈癌患者手术切除的癌组织和80例子宫平滑肌瘤患者经手术切除的正常宫颈组织,采用免疫组化法检测NDC80蛋白的表达水平.qRT-PCR检测正常宫颈细胞和宫颈癌细胞中NDC80 mRNA的表达.将NDC80小干扰RNA(NDC80-siRNA)及其阴性对照(si-NC)分别转染至宫颈癌细胞系中,CCK-8法检测宫颈癌细胞增殖能力,细胞划痕实验检测宫颈癌细胞迁移能力,Transwell实验检测宫颈癌细胞侵袭能力,Western blot检测宫颈癌细胞p-PI3K和p-Akt蛋白表达水平.结果 GEPIA数据库分析显示,NDC80在宫颈癌、膀胱尿路上皮癌、乳腺癌、头颈癌及肺癌等多种肿瘤中呈高表达(P<0.05).NDC80在宫颈癌组织中的表达水平显著高于正常宫颈组织(P<0.05);NDC80在SiHa、CaSki、HeLa、C-33A宫颈癌细胞系中的表达均高于正常宫颈上皮细胞,其中在SiHa与CaSki细胞中表达最显著(P<0.05);敲低NDC80可显著抑制宫颈癌细胞的增殖、迁移及侵袭(P<0.05),同时抑制p-PI3K和p-Akt蛋白的表达水平(P<0.05).结论 在宫颈癌组织中NDC80呈高表达,其可促进宫颈癌细胞的增殖、迁移和侵袭,这一作用可能通过靶向PI3K/Akt信号通路实现.
Objective To investigate the expression of NDC80 in cervical cancer and its mechanism in regulating the proliferation,migration,and invasion of cervical cancer cells.Methods The NDC80 expression in cervical cancer and other multiple tumors was analyzed using the GEPIA database.Cancer tissues were collected from 80 cervical cancer patients who underwent surgical resection at the Third Hospital of Shanxi Medical University,along with normal cervical tissues from 80 patients with uterine leiomyoma who underwent surgical resection.The expression levels of NDC80 protein were detected by immunohistochemistry.The expression of NDC80 mRNA in normal cervical cells and cervical cancer cells was detected by qRT-PCR.NDC80 small interfering RNA(NDC80-siRNA)and its negative control(si-NC)were respectively transfected into cervical cancer cell lines.The CCK-8 assay was used to detect the proliferation ability of cervical cancer cells,the wound healing assay was used to detect the migration ability of cervical cancer cells,the Transwell assay was used to detect the invasion ability of cervical cancer cells,and Western blot was used to detect the expression levels of p-PI3K and p-Akt proteins in cervical cancer cells.Results GEPIA database analysis showed that NDC80 was highly expressed in various types of tumors,including cervical cancer,bladder urothelial carcinoma,breast cancer,head and neck cancer,and lung cancer(P<0.05).The cervical cancer tissues showed significantly higher expression level of NDC80 than that in normal cervical tissues(P<0.05).The cervical cancer cell lines SiHa,CaSki,HeLa,and C-33A showed higher expression of NDC80 than that in normal cervical epithelial cells,with the most significant expression observed in SiHa and CaSki cells(P<0.05).Knockdown of NDC80 significantly inhibited the proliferation,migration,and invasion of cervical cancer cells,and also suppressed the expression levels of p-PI3K and p-Akt proteins(P<0.05).Conclusion NDC80 is highly expressed in cervical cancer tissues and may promote the proliferation,migration and invasion of cervical cancer cells.This effect may be mediated by targeting the PI3K/Akt signaling pathway.
