基于网络药理学和分子对接探讨金银花-鱼腥草药对防治牛乳房炎的作用机制OA
Mechanism of Action of Flos Lonicerae and Cordate Houttuynia Herba Pair in Preventing and Treating Bovine Mastitis Based on Network Pharmacology and Molecular Docking
本研究通过网络药理学和分子对接技术分析金银花-鱼腥草药对防治牛乳房炎的潜在机制,为金银花-鱼腥草药对防治牛乳房炎作用机制研究及其临床应用提供理论依据.在中药系统药理学分析平台数据库中检索分析金银花-鱼腥草药对的活性成分以及所对应的靶点;借助GeneCards、NCBI和GEO等数据库检索牛乳房炎靶点;采用Cytoscape 3.9.1软件和STRING数据库构建"药物-靶点-疾病"及蛋白-蛋白相互作用(PPI)网络图,并筛选金银花-鱼腥草药对防治乳房炎疾病的核心成分和关键靶点;通过DAVID数据库对检索出的潜在靶点进行GO和KEGG等富集分析;利用AutoDock vina软件进行分子对接.结果表明,从金银花-鱼腥草药对中筛选得到有效成分28种,对应靶点192个;筛选得到616个与牛乳房炎相关的靶点;获得金银花-鱼腥草药对防治牛乳房炎潜在靶点143个.从金银花-鱼腥草药对中筛选获得槲皮素、木犀草素、山奈酚、β-谷甾醇、豆甾醇和伞房花耳草素6个核心成分,以及肿瘤坏死因子(TNF)、基质金属蛋白酶9(MMP-9)、白细胞介素-1β(IL1B)、前列腺素内过氧化物合酶 2(PTGS2)、干扰素γ(IFNG)、C-C基序趋化因子配体2(CCL2)、细胞间黏附分子1(ICAM1)和CXC 基序趋化因子配体 8(CXCL8)等防治乳房炎的8个关键靶点.富集分析得到TNF、核因子κB(NF-κB)、白细胞介素-17(IL-17)、核苷酸寡聚化结构域(NOD)样受体和Toll样受体等信号通路.分子对接结果显示,核心成分和关键靶点之间的结合能均小于-5 kcal/mol,表明两者之间能够稳定结合.综上所述,金银花-鱼腥草药对中的活性成分可作用于多靶点来发挥防治牛乳房炎的作用.
This study aimed to analyze the potential mechanism of the Flos lonicerae and Cordate houttuynia herba pair in preventing and treating bovine mastitis using network pharmacology and molecular docking technology,thereby providing a theoretical basis for research on its mechanism of action and clinical application.The active ingredients and corresponding targets of the Flos lonicerae and Cordate houttuynia herba pair were retrieved from the traditional Chinese medicine systems pharmacology database and analysis platform.Targets related to bovine mastitis were ob-tained from databases such as GeneCards,NCBI and GEO.Cytoscape 3.9.1 software and the STRING database were used to construct"drug-target-disease"and protein-protein interaction(PPI)networks,and to screen the core com-ponents and key targets of the herb pair in preventing and treating mastitis.The DAVID database was employed for Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of potential tar-gets.Molecular docking was performed using AutoDock vina software.The results showed that 28 active ingredients and 192 corresponding targets were identified from the herb pair,along with 616 targets associated with bovine mas-titis.A total of 143 potential targets of the herb pair for preventing and treating bovine mastitis were obtained.Among them,6 core components were screened,including quercetin,luteolin,kaempferol,β-sitosterol,stigmasterol and corymbosin;8 key targets for mastitis prevention and treatment were identified,such as tumor necrosis factor(TNF),matrix metalloproteinase 9(MMP-9),interleukin-1β(IL1B),prostaglandin-endoperoxide synthase 2(PTGS2),interferon-gamma(IFNG),C-C motif chemokine ligand 2(CCL2),intercellular adhesion molecule 1(ICAM1)and CXC motif chemokine ligand 8(CXCL8).Enrichment analysis revealed signaling pathways including TNF,nuclear factor-κB(NF-κB),interleukin-17(IL-17),nucleotide-binding oligomerization domain(NOD)-like receptors and toll-like receptor.Molecular docking results demonstrated that the binding energies between core com-ponents and key targets were all<-5 kcal/mol,indicating stable interactions between them.In conclusion,the active ingredients in the Flos lonicerae and Cordate houttuynia herba pair may exert effects on preventing and treating mas-titis through multiple targets.
