次级毛囊的退行本质是毛囊细胞凋亡的过程,本研究从蛋白质组学层面探究该过程的相关蛋白表达,为初步揭示内蒙古绒山羊皮肤次级毛囊凋亡系统的分子调控机制奠定基础.本研究采用SWATH技术,通过比较内蒙古绒山羊9月份(生长旺盛期)与1月份(退行期)的皮肤样本,筛选次级毛囊退行过程中的差异蛋白,并对差异蛋白进行生物信息学分析.结果显示,1%FDR(假阳性)条件下,检测到退行期表达蛋白1 792个,生长期表达蛋白2 199个;筛选出164个退行期特异表达蛋白.并以P<0.05、FC(差异倍数)>2或FC<0.5作为阈值,筛选出内蒙古绒山羊皮肤毛囊退行期与生长期的差异蛋白504个,其中退行期上调蛋白250个,下调蛋白254个.退行期特异表达蛋白主要参与生物学过程(Biological process,BP)的氧化还原过程(Oxidation-reduction process)等功能;在细胞组分(Cellular component,CC)中,它们主要富集于胞外外泌体(Extracellular exosome)中;在分子功能(Molecular function,MF)中,它们主要参与poly(A)RNA结合等功能.差异蛋白在BP中,主要参与翻译(Translation)等功能;在CC中,主要富集于胞外外泌体中;在MF中,参与poly(A)RNA结合功能.KEGG富集分析结果表明,退行期特异表达蛋白与差异蛋白均主要富集于代谢通路(Metabolic pathway),且退行期中富集于TGF-beta信号通路的上调蛋白有PPP2CB、THBS1、Rhoa、TGFB1、Skp1等,下调的蛋白有DCN.从PPI网络中筛选出28个枢纽蛋白,其中16个为核糖体蛋白,占比达57%.综上,TGF-beta信号通路和核糖体蛋白在毛囊退行中起关键调控作用.

The regression of secondary hair follicles is an essential process of follicular cell apoptosis.This study inves-tigated the protein expression related to this process at the proteomics level,laying the foundation for preliminarily re-vealing the molecular regulatory mechanism of the apoptosis system in secondary hair follicles of Inner Mongolia cash-mere goat skin.This study employed SWATH technology to compare skin samples of Inner Mongolia cashmere goat from September(anagen)and January(catagen),screening for differentially expressed proteins during the regression of secondary hair follicles and conducting bioinformatics analysis of these proteins.The results showed that under a 1%FDR(false discovery rate)condition,1 792 proteins were detected in catagen,while 2 199 proteins were detected in anagen.A total of 164 proteins were specifically expressed in catagen.Using a P value<0.05 and FC(fold change)>2 or FC<0.5 as thresholds,504 differentially expressed proteins were identified between catagen and anagen of Inner Mon-golia cashmere goat skin hair follicles,among which 250 were upregulated and 254 were downregulated in catagen.The specifically expressed proteins in catagen were primarily involved in biological processes(BP)such as the oxidation-reduction process;in cellular components(CC),they were mainly enriched in extracellular exosomes;and in molecular functions(MF),they were primarily associated with functions such as poly(A)RNA binding.The differen-tially expressed proteins were mainly involved in BP functions such as translation;in CC,they were predominantly en-riched in extracellular exosomes;and in MF,they participated in poly(A)RNA binding.KEGG enrichment analysis in-dicated that both the specifically expressed protein and the differentially expressed protein in catagen were mainly en-riched in metabolic pathways.Additionally,proteins upregulated in catagen and enriched in the TGF-beta signaling pathway were included PPP2CB,THBS1,Rhoa,TGFB1,Skp1,etc.,while the downregulated protein was DCN.A total of 28 hub proteins were identified from the PPI network,16 of which were ribosomal proteins,accounting for 57%.In summary,the TGF-beta signaling pathway and ribosomal proteins play key regulatory roles in hair follicle catagen.
