电针干预改善高脂饮食诱导肥胖雄性大鼠下丘脑-垂体-性腺轴功能及睾酮合成的机制研究OA
Electroacupuncture improves hypothalamic-pituitary-gonadal axis function and testosterone synthesis in high-fat diet-induced obese male rats
目的:观察电针对高脂饮食诱导肥胖雄性大鼠下丘脑-垂体-性腺(HPG)功能及睾酮合成关键酶的影响,探讨电针改善肥胖相关生殖功能障碍的分子机制.方法:雄性 Wistar大鼠随机分为空白组、模型组、电针组和非穴组,每组8只.高脂饮食8周建立肥胖模型后,电针组予"足三里""肾俞"及"三阴交"干预4周(30 min/次,5次/周),非穴组取电针组对应穴位旁开5 mm处电针.检测各组大鼠体质量、体质量指数(BMI)、Lee's 指 数;ELISA 法检测血清促性腺激素释放激素(GnRH)、黄体生成素(LH)、卵泡刺激素(FSH)、睾酮、雌二醇(E2)水平,ELISA和酶法检测血脂代谢指标;检测附睾精子浓度、活力,计算精子畸形率;HE染色观察睾丸形态学变化;Western blot和 PCR法检测睾丸睾酮合成关键酶类固醇激素调节急性蛋白(StAR)、细胞色素P450侧链裂解酶(P450scc)、17α-羟化酶/17,20-裂解酶(CYP17A)、3β-羟类固醇脱氢酶(3β-HSD)、17β-羟类固醇脱氢酶(17β-HSD)的蛋白和 mRNA表达.结果:与空白组比较,模型组大鼠体质量、BMI及 Lee's指数显著升高(P<0.05),血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)及氧化低密度脂蛋白(OxLDL)含量升高(P<0.05),高密度脂蛋白胆固醇(HDL-C)含量降低(P<0.05),GnRH、睾酮、FSH 和 LH 含量降低(P<0.05),E2 含量升高(P<0.05),精子浓度和活力降低(P<0.05),畸形率升高(P<0.05),睾丸生精小管萎缩,生精细胞排列紊乱,睾酮合成关键酶蛋白和mRNA表达降低(P<0.05).与模型组和非穴组比较,电针组上述指标均显著改善(P<0.05).非穴组除StAR蛋白表达增加外,其他指标与模型组差异无统计学意义.结论:电针能够有效改善肥胖雄性大鼠的生殖功能障碍,其机制可能与恢复HPG轴功能平衡、上调睾酮合成关键酶表达、促进睾酮生成有关.
Objective To observe the effect of electroacupuncture(EA)on hypothalamic-pituitary-gonadal(HPG)axis function and the expression of key enzymes for testosterone synthesis in high-fat diet-induced obese male rats,so as to explore its mechanism in improving obesity-related reproductive dysfunction.Methods Thirty-two 6-week-old male Wistar rats were randomly divided into control,model,EA and non-acupoint EA groups(n=8 in each group).The obesity model was established through high-fat diet feeding for 8 weeks.EA was applied to"Zusanli"(ST36),"Shenshu"(BL23),and"Sanyinjiao"(SP6)for 30 min,5 d a week for 4 weeks.The body weight,body mass index(BMI)and Lee's index of rats were detected.The levels of serum gonadotropin-releasing hormone(GnRH),luteinizing hormone(LH),follicle-stimulating hormone(FSH),testosterone,estradiol(E2)and lipid metabolism indexes were detected by ELISA and enzymic method.The epididymal sperm concentration and motility were assessed,and the sperm deformity rate was calculated.The morphological changes of testis were observed by HE staining.The protein and mRNA expression of steroid-regulated acute protein(StAR),cytochrome P450 side chain cleavage enzyme(P450scc),17α-hydroxylase/17,20-lyase(CYP17A),3β-hydroxysteroid dehydrogenase(3β-HSD)and 17β-hydroxysteroid dehydrogenase(17β-HSD)were detected by Western blot and PCR,respectively.Results Compared with the control group,the body weight,BMI,Lee's index,the levels of serum TC,TG,LDL-C,OxLDL and E2,the deformity rate were significantly increased(P<0.05)in the model group,while the levels of HDL-C,GnRH,testosterone,FSH and LH,the sperm concentration and motility,the protein and mRNA expression of key enzymes in testosterone synthesis were significantly decreased(P<0.05).Compared with the model and the non-acupoint EA groups,the indexes mentioned above were significantly improved in the EA group(P<0.05).Except for increased StAR protein expression,no significant differences were found in other indicators between the non-acupoint EA group and the model group.Conclusion EA can effectively improve reproductive dysfunction in obese male rats.The mechanism may be related to restoring the functional balance of the HPG axis,up-regulating the expressions of key testosterone synthesis enzymes,and promoting testosterone production.
