基于二仙孕子丸治疗多囊卵巢综合征所致不孕症作用机制的网络药理学和分子对接分析OA
Network Pharmacology and Molecular Docking Analysis of the Mechanism of Erxian Yunzi Pills in the Treatment of Infertility Caused by Polycystic Ovary Syndrome
目的 采用网络药理学和分子对接技术,探讨二仙孕子丸治疗多囊卵巢综合征所致不孕症的作用及分子机制.方法 从TCMSP数据库中筛选出二仙孕子丸中的主要成分,并使用UniProt 数据库获取其潜在靶点.通过GeneCard数据库和OMIM数据库检索和筛选多囊卵巢综合征所致不孕症相关靶标.将药物和疾病靶点取交集,通过STRING数据库获取蛋白质-蛋白质相互作用(PPI)网络,并使用Cytoscape3.7.2 软件进行可视化分析.利用R软件函数包分析通过GO和KEGG通路富集分析确定的共同靶点的潜在机制,并采用微生信对结果进行可视化.使用AutoDockVina1.1.2 和PyMoL软件对二仙孕子丸主要活性化合物与多囊卵巢综合征所致不孕症关键靶点进行分子对接验证.结果 共发现二仙孕子丸有 218 种化合物,二仙孕子丸治疗多囊卵巢综合征所致不孕症的潜在靶点 201 个,核心靶点 22 个,基于信号通路分析得治疗多囊卵巢综合征所致不孕症密切相关核心关键靶点 5 个,分别为TP53、IL6、AKT1、TNF和STAT3.GO功能富集分析结果表明二仙孕子丸主要参与多种细胞应激反应的生物过程.KEGG信号通路富集分析潜在靶点在炎症、氧化应激及癌症通路高度富集.分子对接验证二仙孕子丸活性成分槲皮素、木犀草素、山柰酚、卢皮醇与 TP53、IL6、AKT1、TNF 和 STAT3 均对接稳定,其中槲皮素与 AKT1 结合性最好.结论 二仙孕子丸对多囊卵巢综合征所致不孕症的治疗效果是生物活性成分的多靶点和多个信号通路的作用,本研究的发现可为后续的基础实验验证和新的研究方向铺平道路.
Objective To explore the effect and molecular mechanism of Erxian Yunzi pills in the treatment of infertility caused by polycystic ovary syndrome by network pharmacology and molecular docking technology.Methods The main components of Erxian Yunzi pills were screened from the TCMSP database,and the potential targets were obtained by using the UniProt database.The GeneCard database and OMIM database were used to search and screen the targets related to infertility caused by polycystic ovary syndrome.The drug and disease targets were intersected,and the protein-protein interaction(PPI)network was obtained through the String database,and Cytoscape3.7.2 software was used for visual analysis.The potential mechanism of common targets identified by GO and KEGG pathway enrichment analysis was analyzed by R software function package,and the results were visualized by microbiome.AutoDockVina1.1.2 and PyMoL software were used to verify the molecular docking between the main active compounds of Erxian Yunzi pills and the key targets of infertility caused by polycystic ovary syndrome.Results A total of 218 compounds were found in Erxian Yunzi pills.There were 201 potential targets and 22 core targets for Erxian Yunzi pills in the treatment of infertility caused by polycystic ovary syndrome.Based on signal pathway analysis,there were 5 core key targets closely related to the treatment of infertility caused by polycystic ovary syndrome,which were TP53,IL6,AKT1,TNF and STAT3.The results of GO functional enrichment analysis showed that Erxian Yunzi pills was mainly involved in the biological processes of various cellular stress responses.KEGG signaling pathway enrichment analysis showed that potential targets were highly enriched in inflammation,oxidative stress and cancer pathways.Molecular docking verified that the active components of Erxian Yunzi Pills,quercetin,luteolin,kaempferol,lupeol,were docked stably with TP53,IL6,AKT1,TNF and STAT3,among which quercetin had the best binding to AKT1.Conclusion The therapeutic effect of Erxian Yunzi pills on infertility caused by polycystic ovary syndrome is the effect of multiple targets and multiple signaling pathways of bioactive components.The findings of this study can pave the way for subsequent basic experimental verification and new research directions.
