基于网络药理学探讨灵芝抗心力衰竭炎症反应的作用机制OA
Mechanism of action of Lingzhi(Ganoderma)against inflammatory response in heart failure based on network pharmacology
目的 探讨"药食同源"代表性中药灵芝干预心力衰竭炎症反应的潜在活性小分子及其作用机制.方法 整合中药系统药理学数据库与分析平台(TCMSP)等与基因表达综合数据库(GEO)筛选灵芝候选活性成分及心衰差异靶标.借助Cytoscape 3.10.4 软件构建网络并识别关键靶点.应用R语言进行功能富集分析,运用CB-Dock2 执行分子对接验证.结果 槲皮素、染料木黄酮等小分子成分显示出较高的网络连接度,基质金属蛋白酶(MMP)-9、白细胞介素(IL)-1β基因等 71 个交集靶点主要富集于炎症反应调控及IL-17、Toll样受体(TLR)等通路;候选成分中还识别出Ganoderic acid F等灵芝特征性三萜.代表性黄酮类成分与MMP-9、IL-1β结合自由能均≤-5.0 kcal/mol,并与结合腔内多个残基形成氢键、疏水相互作用等非共价相互作用.结论 在当前数据库可识别的小分子成分框架下,灵芝可能通过作用于IL-1β、MMP-9 等靶点参与心衰炎症的调控,但上述结果不能直接外推为灵芝全成分体系的整体作用机制.
Objective To investigate the potential active small molecules and their mechanisms of action of Lingzhi(Ganoderma),a representative Chinese medicinal herb with food-medicine homology,in intervening in the inflammatory response of heart failure.Methods Databases including TCMSP and GEO chip data were integrated to screen candidate active components of Lingzhi(Ganoderma)and differential targets associated with heart failure.The network was constructed using Cytoscape 3.10.4 software,and key targets were identified.Functional enrichment analysis was performed using R language,and molecular docking validation was carried out using CB-Dock2.Results Small-molecule components such as quercetin and genistein exhibited relatively high network connectivity.A total of 71 intersecting targets,including matrix metalloproteinase(MMP)-9 and interleukin(IL)-1β,were mainly enriched in pathways related to the regulation of inflammatory response,IL-17,and Toll-like receptor(TLR)signaling.Among the candidate components,Lingzhi(Ganoderma)characteristic triterpenes such as ganoderic acid F were also identified.Representative flavonoid components exhibited binding free energies≤-5.0 kcal/mol with MMP-9 and IL-1β,and formed non-covalent interactions such as hydrogen bonds and hydrophobic interactions with multiple residues in the binding pocket.Conclusion Within the framework of small-molecule components identifiable from current databases,Lingzhi(Ganoderma)may participate in the regulation of heart failure inflammation by acting on targets such as IL-1β and MMP-9.However,these findings cannot be directly extrapolated to the overall mechanism of action of the whole component system of Lingzhi(Ganoderma).
