目的 运用网络药理学方法探讨安乐阴方外治外阴阴道假丝酵母菌病(VVC)的作用机制.方法 利用TCMSP、PubChem、Swiss ADME 及 Swiss Target Prediction 等数据库筛选出安乐阴方主要化学成分及其潜在靶点,Gen-eCards、OMIM 数据库获得 VVC 相关基因并交集;利用 STRING 数据库构建蛋白质相互作用(PPI)网络;通过 Cyto-scape 3.10.3 构建药物复方-中药-成分-靶点的网络图;借助微生信-在线生物信息学分析、可视化云平台进行基因本体(GO)、京都基因与基因组百科全书(KEGG)富集分析;通过 CB-Dock2 网站进行分子对接验证.结果 网络药理学分析筛选得到安乐阴方中有效化学成分94 个,药物靶点954 个,VVC 相关靶基因2924 个,药物与疾病共同交集靶点429 个.最终确定AKT1、GAPDH、TP53、TNF、STAT3 等5 个核心靶点,安乐阴方可能通过癌症中的蛋白聚糖、脂质与动脉粥样硬化等信号通路(包括癌症中的蛋白聚糖、脂质与动脉粥样硬化通路),减少细胞凋亡、调控自噬与细胞黏附,影响细胞增殖分化,并发挥抗炎作用从而治疗 VVC.分子对接结果显示,安乐阴方中香叶木素、茵芋碱、花椒素 N 与 AKT1、GAPDH、TP53 具有一定的亲和力.结论 安乐阴方可能通过多种活性成分、多个靶点及多条信号通路发挥对 VVC 的治疗作用.

Objective To investigate the mechanism of action of Anle Yin Formula in the external treatment of vulvovagi-nal candidiasis(VVC)using network pharmacology.Methods The main chemical constituents and their potential targets of Anle Yin Formula were identified through databases including TCMSP,PubChem,Swiss ADME,and Swiss Target Predic-tion.VVC-related genes were retrieved from the GeneCards and OMIM databases,and the intersection between the two sets was determined.A protein-protein interaction(PPI)network was constructed using the STRING database.A network dia-gram of drug compound-traditional Chinese medicine-component-target was created through Cytoscape 3.10.3.Gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis were conducted via the Weishengxin cloud platforman—an online tool for bioinformatics analysis and visualization.Molecular docking validation was performed using the CB-Dock2 website.Results Through network pharmacology analysis,94 active chemical components in Anle Yin Formula,954 drug targets,and 2924 VVC-related target genes were identified,with 429 overlapping targets between the drug and the disease.Five core targets—AKT1,GAPDH,TP53,TNF,and STAT3—were ultimately determined.This sug-gested that Anle Yin Formula might reduce cell apoptosis,regulate autophagy and cell adhesion,and affect cell proliferation and differentiation through signaling pathways such as proteoglycan,lipid,and atherosclerosis in cancer(including pathways related to proteoglycan,lipid,and atherosclerosis in cancer),and thereby exerting anti-inflammatory effects to treat VVC.Molecular docking results indicated that Diosmetin,Skimmianin and Xanthoxylin N in Anle Yin Formula exhibited certain af-finity for AKT1,GAPDH,and TP53.Conclusion Anle Yin Formula may exert therapeutic effects on VVC through multi-ple active ingredients,multiple targets,and multiple signaling pathways.
