为证明基于RTILs的SEM预处理方法对湿软生物样品的有用性.本研究以RTILs简单滴加与吸干处理替代常规费时、繁琐及费用高的SEM预处理方法.结果表明,基于RTILs的SEM预处理法虽在保持样品形态方面不是非常完美,但与常规方法所得结果接近,且在孢子数量、孢子链、叶毛以及叶片表皮细胞等形态保持方面比常规SEM预处理法更接近于真实情况,且导电性能可媲美常规预处理法.所构建的基于RTILs的SEM预处理法在替代常规SEM预处理法方面存在巨大潜力,其具有经济、简便等特点,且所得图片质量与常规预处理法几乎一致,尤其在保持链格孢菌孢子串珠状形态、细胞饱满度等方面优于常规预处理法.

To demonstrate the utility of the RTIL-based SEM pretreatment method for hydrated soft biological specimens,this study replaced the conventional time-consuming,labor-intensive,and expen-sive SEM sample preparation with a simple droplet application and blotting treatment.The results showed that although the RTIL-based method did not achieve perfect preservation of morphology,it produced results comparable to those of conventional methods.Specifically,it demonstrated superior structural fidelity in preserving spore quantity,spore chains,leaf trichomes,and epidermal cells,provi-ding a more realistic representation of biological structures than traditional SEM pre-treatment.Addi-tionally,its electrical conductivity was comparable to that of conventional methods.The developed RTIL-based approach shows great potential as an alternative to conventional SEM pretreatment,offer-ing advantages in cost,simplicity,and efficiency,while maintaining image quality nearly identical to that of traditional protocols.Notably,it outperforms standard methods in preserving critical structural features such as the bead-like morphology of Alternaria spore chains and cellular turgor.
