基于环境DNA技术的海河干流浮游动植物资源调查与分析OA

为了探究不同人类活动干扰强度下海河干流浮游动植物的物种组成和群落结构特征,本研究采用环境DNA技术,结合18S rRNA高通量测序和生物信息学分析等方法,以海河干流12个位点采集的水体样本为研究对象,调查该区域的浮游动植物资源情况,并分析高、中、低3种人为干扰强度对海河干流浮游动植物的影响.结果显示:①检测出的浮游动物分别隶属于7门56种,排名前3的门类为纤毛门(Ciliophora)、轮虫动物门(Rotifera)和节肢动物门(Arthropoda),其中纤毛门占绝对优势,各位点间共有的浮游动物优势种为Orborhabdostyla bromelicola、淡水筒壳虫(Tintinnidium fluviatile)和棕色钟虫(Vorticella fusca),整个调查区域相对丰度排名第一的优势种为中华哲水蚤(Sinocalanus sinensis);②检测出的浮游植物分别隶属于4门52种,排名前3的门类为硅藻门(Bacillariophyta)、绿藻门(Chlorophyta)和隐藻门(Cryptophyta),各位点间共有的浮游植物优势种为Uncultured Cryptomonas、梅尼小环藻(Cyclotella meneghiniana)、弯隐藻(Cryptomonas curvata)等;③在群落结构多样性方面,浮游动物的Alpha多样性指数和Beta多样性指数在不同强度干扰组间的差异均不显著(P>0.05),浮游植物的Alpha多样性指数虽然组间差异不显著(P>0.05),但Beta多样性指数的组间差异显著(P<0.05),意味着在面对人类活动干扰时浮游植物比浮游动物更为敏感;④本次调查中,浮游植物中硅藻占优势地位且未检出蓝藻门,浮游动物中中华哲水蚤为优势种且轮虫的多样性和相对丰度较低,这些结果表明海河干流的整体水质状况良好.

To investigate the species composition and community structure of zooplankton and phytoplankton in the Haihe River mainstream under different intensities of human activity disturbance,water samples were collected from 12 sites along the Haihe River mainstream.The situation of plankton resources in this area was investigated using environmental DNA(eD-NA)technology combined with 18S rRNA gene highthroughput sequencing and bioinformatic analysis.Meanwhile,the impacts of high,medium,and low levels of anthropogenic disturbance on zooplankton and phytoplankton were analyzed.The results illustrated that:① A total of 56 species belonging to seven phyla were detected among zooplankton,and the top three phyla were Ciliophora,Rotifera,and Arthropoda,among which Ciliophora was absolutely predominant.The dominant zoo-plankton species common across all sampling sites were Orborhabdostyla bromelicola,Tintinnidium fluviatile,and Vorticella fusca,and Sinocalanus sinensis was the dominant species with the highest relative abundance in the entire survey area.② A total of 52 species belonging to four phyla of phytoplankton were detected,and the top three phyla were Bacillariophyta,Chlorophyta,and Cryptophyta.The dominant phytoplankton species common across all sites were Uncultured Cryptomonas,Cyclotella meneghiniana,Cryptomonas curvata,etc.③ In terms of the diversity of the community structure,the differences in the Alpha diversity index and Beta diversity index of zooplankton were not significant(P>0.05)among the groups with dif-ferent intensities of human activity disturbance,while the Alpha diversity index of phytoplankton was not significant(P>0.05),but the difference in the Beta diversity index was significant(P<0.05)among different groups,which meant that phytoplankton was more sensitive than zooplankton when facing anthropogenic disturbance.④ Bacillariophyta was dominant in phytoplankton and the species belonging to Cyanophyta were not detected,and Sinocalanus sinensis was the dominant species in zooplankton with a low diversity and relative abundance of Rotifers,which indicates a good overall water quality condition of the Haihe River mainstresm.
