不同激素对甜瓜未授粉子房愈伤组织诱导的影响OA

Effects of Phytohormones on Callus Induction from Unpollinated Ovaries Culture of Melon

为系统探讨不同植物激素及其组合对甜瓜未授粉子房愈伤组织诱导的影响,通过设置不同浓度的 GA3、NAA、KT、2,4-D、6-BA 和 TDZ 单一处理及组合处理,并在培养基中加入不同浓度椰汁,对不同处理下甜瓜未授粉子房愈伤组织诱导率及愈伤形态特征进行分析.结果表明,在 GA3、TDZ、NAA、KT 这4种激素的正交试验组合中,对甜瓜愈伤诱导的效果依次为 NAA>KT>GA3>TDZ,最优水平分别为0.5 mg·L-1NAA、0.1 mg·L-1KT、0.5 mg·L-1GA3 和 0.06 mg·L-1TDZ.0.5 mg·L-1NAA 与 0.1 mg·L-1KT 组合处理对甜瓜未授粉子房愈伤组织的诱导效果最佳,愈伤组织形成率为 52.6%,且质地致密、呈淡绿色;在 MS+0.5 mg·L-1NAA+0.1 mg·L-1KT 培养基中添加 15%椰汁后,愈伤诱导率提升至68.1%,褐化率降至7.8%.单一激素 TDZ、2,4-D 和6-BA 易导致愈伤组织褐化或畸形生长.综上,诱导甜瓜未授粉子房愈伤组织的高效培养基组合为 MS+0.5 mg·L-1NAA+0.1 mg·L-1KT+15%椰汁.

To systematically investigate the effects of different phytohormones and their combinations on the callus induction from in vitro unpollinated ovary culture of melon,different concentrations of GA3,NAA,KT,2,4-D,6-BA and TDZ in individually and in combined treatments were designed,and different concentrations of coconut milk were incorporated into the medium,the effects of these factors on the callus induction rate and the morphological characteristics of callus were analyzed.The results showed that among the different combinations of GA3,TDZ,NAA,and KT in the orthogonal experiment,the order of their effects on the callus induction was NAA>KT>GA3>TDZ.The optimal concentrations were 0.5 mg·L-1NAA,0.1 mg·L-1KT,0.5 mg·L-1GA3 and 0.06 mg·L-1TDZ re-spectively.The combination of 0.5 mg·L-1NAA and 0.1 mg·L-1 KT had the best effect on callus induction,with an induction rate of 52.6%,producing callus with a dense texture and light green color.In the optimised medium containing MS+0.5 mg·L-1NAA+0.1 mg·L-1KT with the addition of 15%coconut milk,the rate of callus induction was enhanced to 68.1%,and browning rate was reduced to 7.8%.Single hormones TDZ,2,4-D,6-BA was prone to cause browning or abnormal growth of callus.In conclusion,the highly effi-cient medium for callus induction from in vitro unpollinated ovary culture of melon is MS medium supplemented with 0.5 mg·L-1 NAA,0.1 mg·L-1 KT,and 15%coconut milk.
