地黄对急性痛风性关节炎的预防作用及其机制研究OA

目的 研究地黄提取物干预大鼠急性痛风性关节炎(AGA)的预防效果与作用机制.方法 建立Wistar大鼠AGA动物模型,观察造模后、模型组和给药组大鼠以及空白组的踝关节肿胀程度指数的变化,对比造模前后各组大鼠体质量变化指数、肾脏指数以及肝脏指数;检测大鼠血清中尿酸;肌酐含量和黄嘌呤氧化酶的活性以及IL-1β、NLRP3、TNF-α的含量;采用HE染色观察踝关节组织病理学形态变化;通过UPLC-TSQ-MS技术对地黄水提物浸膏和乙酸乙酯萃取物浸膏进行成分分析.结果 相对模型组,给药组大鼠表现出较好的抗关节炎活性,血清中尿酸、肌酐IL-1β、NLRP3、TNF-α含量以及黄嘌呤氧化酶活力等指标均明显降低;UPLC-TSQ-MS分析检测出地黄水提物浸膏共18种成分,地黄乙酸乙酯萃取物浸膏共14种成分.结论 地黄水提取物浸膏和乙酸乙酯萃取物浸膏均对急性痛风性关节炎有较好的防治作用,其作用机制可能是通过抑制炎症因子的释放,降低尿酸的形成,抑制黄嘌呤氧化酶的活性以及促进肌酐的排泄等发挥预防AGA的作用.

Objective To study the preventive effect and potential mechanism of Rhizoma Rehmanniae extract against acute gouty arthritis(AGA)in rats.Methods Wistar rats were used to establish an AGA rat model.Then,the ankle joint swelling index was observed,and changes in body weight index,kidney index,and liver index were recorded before and after modeling.Serum levels of uric acid,creatinine,xanthine oxidase activity,and the contents of IL-1β,NLRP3,and TNF-α were measured.HE staining was used to observe pathological changes in the ankle joint tissue.Additionally,UPLC-TSQ-MS was employed to analyze the components of Rhi-zoma Rehmanniae aqueous extract and its ethyl acetate extract paste.Results Compared with the model group,the treatment group dem-onstrated good activity against AGA.The levels of uric acid,creatinine,IL-1β,NLRP3,TNF-α,and the xanthine oxidase activity in serum were significantly reduced.Furthermore,UPLC-TSQ-MS analysis showed 18 components in the aqueous extract paste of Rhizoma Rehmanniae and 14 components in the ethyl acetate extract paste.Conclusion Both the aqueous extract and ethyl acetate extract paste of Rhizoma Rehmanniae have good preventive effects on AGA.The mechanism of the action maybe involve inhibiting the release of inflam-matory factors,reducing uric acid production,suppressing xanthine oxidase activity,and promoting creatinine excretion,thereby exert-ing preventive effects against AGA.
