当归补血汤干预斑秃的机制研究OA

Effects of Danggui Buxue Decoction(当归补血汤)on follicular cell apoptosis and expression of hair-related growth factors in imiquimod-induced alopecia areata mouse models

目的 通过观察当归补血汤对小鼠斑秃模型血清及皮损碱性磷酸酶(ALP)、胰岛素样生长因子(IGF)和成纤维细胞生长因子-7(FGF-7)表达的影响,初步探讨当归补血汤干预斑秃可能的作用机制.方法 将造模成功小鼠随机分为模型组、当归补血汤低剂量组(2.5g/kg)、当归补血汤中剂量组(5g/kg)、当归补血汤高剂量组(10g/kg)、维生素D组(30ng/kg).另设正常组生理盐水灌胃,干预45天.光镜下观察皮损区毛发脱落及生长情况;HE染色观察小鼠皮损病理;原位末端标记法(TUNEL)染色检测小鼠皮损毛囊细胞凋亡率;ELISA检测小鼠血清ALP、IGF、FGF-7水平;Real-Time qPCR检测小鼠皮损ALP、IGF、FGF-7 mRNA表达.结果 与正常组比较,模型组皮损区毛发明显脱落,镜下视野毛囊数目减少,毛囊细胞周围呈空泡样改变;各药物干预组皮损区可见新生毛发,当归补血汤高剂量组及维生素D组脱发情况改善明显,部分毛发增长,镜下视野毛囊数目增多,空泡样改变减轻.与正常组比较,模型组毛囊细胞凋亡率显著升高(P<0.05),血清ALP、IGF、FGF-7水平及皮损mRNA表达显著下调(P<0.05);与模型组比较,当归补血汤中、高剂量组和维生素D组毛囊细胞凋亡率显著降低(P<0.05),血清ALP、IGF、FGF-7水平及皮损mRNA表达显著上调(P<0.05).结论 当归补血汤有效干预斑秃可能与调节ALP、IGF、FGF-7表达、减轻毛囊细胞凋亡有关.

Objective To preliminarily investigate the mechanism of Danggui Buxue Decoction(当归补血汤,DGBXD)in treating alo-pecia areata(AA)by examining its effects on serum alkaline phosphatase(ALP),insulin-like growth factor(IGF),fibroblast growth factor-7(FGF-7)levels,and their mRNA expression in skin lesions of AA mouse models.Methods AA mice were induced and ran-domly divided into a model group,low-dose DGBXD(2.5 g/kg),medium-dose DGBXD(5 g/kg),high-dose DGBXD(10 g/kg),and vitamin D group(30 ng/kg),with a normal control group receiving saline.Interventions lasted 45 days.Hair loss and regeneration were observed under light microscopy.Pathological changes were analyzed via HE staining.TUNEL staining quantified follicular cell apopto-sis.Serum ALP,IGF,and FGF-7 levels were measured by ELISA.Real-time qPCR assessed mRNA expression of ALP,IGF,and FGF-7 in skin lesions.Results Compared to the normal group,the model group exhibited severe hair loss,reduced hair follicle density,and vacuolar degeneration.Drug-treated groups,especially high-dose DGBXD and vitamin D,showed hair regrowth,increased fol-licles,and reduced vacuolar changes.The model group had significantly elevated follicular apoptosis rates(P<0.05),decreased serum ALP,IGF,FGF-7 levels,and downregulated mRNA expression(P<0.05).In contrast,medium-and high-dose DGBXD and vitamin D groups demonstrated reduced apoptosis rates(P<0.05)and upregulated ALP,IGF,FGF-7 levels and mRNA expression(P<0.05).Conclusion DGBXD may mitigate alopecia areata by modulating ALP,IGF,and FGF-7 expression and alleviating follicular cell apopto-sis,suggesting a multi-target therapeutic mechanism.
