基于低氧诱导因子-1α防治肾间质纤维化研究进展OA
Research Progress on Therapeutic Approaches for Renal Interstitial Fibrosis and the Pathogenic Role of Hypoxia-Inducible Factor-1α
肾间质纤维化(renal interstial fibrosis,RIF)是慢性肾脏病(chronic kidney disease,CKD)进展的共同途径,以终末期肾病(end-stage renal disease,ESRD)为终点.低氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)是维持氧稳态核转录因子的关键因子,缺氧条件下,被激活的 HIF-α 受多种信号通路的影响作用于 RIF.文章通过综述 HIF-1α 的生物学特性、RIF 发病机制、HIF-1α 诱导 RIF 的信号通路及 RIF 的可能治疗靶点,认为 HIF-1α 在肾间质纤维化过程中起了重要作用,以 HIF-1α 为治疗靶点是一种很有前景的治疗方案,以期对防治 RIF、改善肾功能、延缓 CKD 进展提供一定的思考.
Renal interstitial fibrosis(RIF)is the common pathway of chronic kidney disease(CKD),and end-stage renal disease(ESRD)is the end point.Hypoxia-inducible factor-1α(HIF-1α),a key nuclear transcription factor maintai-ning oxygen homeostasis,is activated under hypoxic conditions.The activated HIF-α influences RIF through multiple signa-ling pathways.The article reviews the biological characteristics of HIF-1α,the pathogenesis of RIF,the signaling pathways by which HIF-1α induces RIF,and potential therapeutic targets for RIF,it proposes that HIF-1α plays a crucial role in the process of renal interstitial fibrosis and suggests that targeting HIF-1α represents a promising therapeutic approach.The aim is to offer insights for preventing and treating RIF,improving renal function,and delaying the progression of CKD.
