目的 探讨川芎嗪对子宫内膜异位症大鼠异位内膜组织生长的抑制作用,并基于血管内皮生长因子A(VEGFA)和肿瘤坏死因子-α(TNF-α)的表达变化探讨其可能的作用机制.方法 采用自体子宫内膜移植法构建子宫内膜异位症SD大鼠模型.将造模成功大鼠随机分为模型组及低、中、高剂量川芎嗪组,每组6只.各给药组分别灌胃给予2.375,4.75,9.5 mg/mL的磷酸川芎嗪溶液(1 mL/d),模型组灌胃等体积生理盐水,均连续28 d.干预后测量异位内膜组织体积并计算体积缩小率;采用HE染色观察内膜组织病理学变化;分别通过免疫组化法异位内膜组织中、ELISA法检测血清及腹腔冲洗液中VEGFA、TNF-α的蛋白水平.结果 与模型组相比,中、高剂量川芎嗪组异位内膜组织体积缩小率显著升高(P<0.05),低、中、高剂量川芎嗪组异位内膜组织体积缩小率剂量依赖性升高(P<0.05).与模型组相比,高剂量川芎嗪组异位内膜组织及血清中VEGFA、TNF-α的表达水平均显著降低(P<0.05);而模型组及低、中、高剂量川芎嗪组腹腔冲洗液中VEGFA、TNF-α水平差异无统计学意义(P>0.05).结论 中、高剂量川芎嗪灌胃可有效抑制大鼠异位内膜组织生长,同时伴随异位内膜组织及血清中VEGFA、TNF-α表达水平的降低,其作用机制可能与抑制血管生成和炎性微环境有关.

Objective To investigate the inhibitory effect of tetramethylpyrazine(TMP)on the growth of ectopic endometrial tissue in rats with endometriosis,and to explore its underlying mechanism by examining the expressions of vascular endothelial growth factor A(VEGFA)and tumor necrosis factor-alpha(TNF-α).Methods An endometriosis model was established in Sprague-Dawley(SD)rats using autologous endometrial implantation.The successfully modeled rats were randomly divided into model group,low-,medium-,and high-dose TMP groups,with 6 rats in each group.The rats in TMP groups were administered with tetramethylpyrazine phosphate solution(1 mL/d)at concentrations of 2.375,4.75,9.5 mg/mL by gavage for 28 consecutive days,respectively,while the rats in model group received an equal volume of normal saline.After the intervention,the volume of ectopic endometrial tissue was mea-sured,and the volume reduction rate was calculated.Histopathological changes were observed using hematoxylin-eosin(HE)staining.The protein levels of VEGFA and TNF-α in ectopic endometrial tissue were detected by immunohis to chemistry,and those in serum and peritoneal lavage fluid were mearured by enzyme-linked immunosorbent assay(ELISA),respectively.Results Compared with model group,the volume reduction rate of ectopic endometrial tissue in medium-and high-dose TMP groups was significantly increased(P<0.05),and the volume reduction rate dose-dependently increased by TMP(P<0.05).Compared with model group,the expression levels of VEGFA and TNF-α in both ectopic endometrial tissue and serum significantly decreased in high-dose TMP group(P<0.05);how-ever,there was no statistically significant difference in the levels of VEGFA and TNF-α in peritoneal lavage fluid among all groups(P>0.05).Conclusion The medium-and high-dose TMP by gavage can effectively inhibit the growth of ectopic endometrial tissue in rats,accompanied by a decrease in the expression levels of VEGFA and TNF-α in both ectopic endometrial tissue and serum.Its mechanism of action may be associated with the inhibition of angiogenesis and the inflammatory microenvironment.
