AIM2对宫颈癌细胞PANoptosis的影响及其机制OA
Effect and mechanism of AIM2 on PANoptosis in cervical cancer cells
目的 探讨AIM2对宫颈癌HeLa细胞PANoptosis的影响及潜在机制.方法 ①采用UCSC数据库分析AIM2基因在宫颈癌中的表达水平.通过Western blot和RT-qPCR检测宫颈癌细胞HeLa和C33a及正常宫颈上皮细胞HCer中AIM2及PANop-tosis相关分子[细胞焦亡(Caspase-1)、凋亡(Caspase-3,cleaved-Caspase-3)、坏死性凋亡(MLKL,p-MLKL)]的表达水平.②通过慢病毒转染构建AIM2稳定敲低的HeLa细胞模型(sh-AIM2),并设空白组和阴性对照组(sh-NC).采用Western blot检测各组细胞中AIM2和PANoptosis相关蛋白的表达;通过EthD-Ⅲ、TUNEL及PI染色分别检测细胞焦亡、凋亡及坏死性凋亡情况.③基于UCSC、GEPIA数据库分析宫颈癌PLA2G7基因表达水平及其与AIM2基因表达的相关性;通过Western blot检测宫颈癌细胞中PLA2G7的基础表达及敲低AIM2后的变化.④将HeLa细胞分为control组、Melittin(PLA2G7激动剂)组、sh-AIM2组、sh-AIM2+Melittin组,Western blot检测各组细胞中AIM2、PLA2G7及PANoptosis相关分子的表达水平.结果 ①UCSC数据库中AIM2在宫颈癌组织的表达显著高于正常宫颈上皮组织(P<0.05).与HCer细胞及C33a细胞相比,HeLa细胞中AIM2的蛋白及mRNA水平均显著升高(P<0.05),Caspase-1、Caspase-3、MLKL的mRNA水平升高(P<0.05);Caspase-1、cleaved-Caspase-3、p-MLKL蛋白表达升高(P<0.05).②与空白组比较,sh-AIM2组细胞中AIM2的蛋白表达水平降低(P<0.05),Caspase-1、cleaved-Caspase-3、p-MLKL的表达均降低(P<0.05),细胞焦亡、凋亡及坏死性凋亡水平均降低(P<0.05).③UCSC数据库中PLA2G7在宫颈癌组织的表达显著高于其在正常宫颈上皮组织的表达(P<0.05),GEPIA数据库显示宫颈癌患者中AIM2与PLA2G7的表达水平存在正相关(r=0.27,P<0.05).Western blot结果显示,与C33a细胞相比,HeLa细胞中PLA2G7显著升高,下调AIM2后PLA2G7表达显著降低(P<0.05).④与control组相比,Melittin组HeLa细胞中PLA2G7、Caspase-1、cleaved-Caspase-3、p-MLKL的蛋白表达上调(P<0.05),而sh-AIM2组上述分子表达则显著下调(P<0.05).与sh-AIM2组相比,sh-AIM2+Melittin组联合处理可逆转上述分子的低表达水平(P<0.05).结论 宫颈癌中AIM2和PLA2G7高表达且呈正相关,下调AIM2可降低PLA2G7表达,二者协同促进宫颈癌HeLa细胞PANoptosis,为宫颈癌治疗提供潜在方向.
Objective To investigate the effect of AIM2 on PANoptosis in cervical cancer HeLa cell and its underlying mechanism.Methods ①The expression level of AIM2 gene in cervical cancer was analyzed using UCSC database.Western blot and RT-qPCR were performed to detect the expressions of AIM2 and PANoptosis-related molecules[pyroptosis(Caspase-1),apoptosis(Caspase-3,cleaved-Caspase-3),necroptosis(MLKL,p-MLKL)]in cervical cancer cells HeLa,C33a and normal cervical epithelial HCer cells.②Lentiviral transfection was used to establish a stable AIM2-knockdown HeLa cell model(sh-AIM2),and blank group and negative control group(sh-NC)were set up in the experiment.Western blot was used to detect the expressions of AIM2 and PANoptosis-related pro-teins.Pyroptosis,apoptosis and necroptosis were detected by EthD-Ⅲ,TUNEL and PI staining,respectively.③The expression level of PLA2G7 gene in cervical cancer cell and its correlation with AIM2 expression were analyzed based on UCSC and GEPIA databases,respectively.Western blot was used to detect the expression of PLA2G7 in cervical cancer cells and its change after AIM2 knockdown.④HeLa cells were divided into control group,Melittin(PLA2G7 agonist)group,sh-AIM2 group,and sh-AIM2+Melittin group.Western blot was used to detect the expressions of AIM2,PLA2G7 and PANoptosis-related molecules.Results ①Based on UCSC database,AIM2 expression was significantly higher in cervical cancer tissues than in normal cervical epithelial tissues(P<0.05).Compared with HCer and C33a cells,HeLa cells exhibited increased protein and mRNA levels of AIM2,elevated mRNA levels of Caspase-1,Caspase-3 and MLKL,and increased protein expressions of Caspase-1,cleaved-Caspase-3 and p-MLKL(all P<0.05).②Compared with blank group,AIM2 protein expression,the expressions of Caspase-1,cleaved-Caspase-3 and p-MLKL,and the levels of pyroptosis,apoptosis and necroptosis were significantly reduced in sh-AIM2 group(P<0.05).③UCSC database analysis results showed that PLA2G7 expression was significantly higher in cervical cancer tissues than in normal cervical epithelial tissues(P<0.05).GEPIA database analysis results showed that there was a positive correlation between AIM2 expression and PLA2G7 expression in cervical cancer patients(r=0.27,P<0.05).Western blot results indicated that PLA2G7 expression was significantly higher in HeLa cells than in C33a cells,and that PLA2G7 expression was significantly decreased after AIM2 down-regulation(P<0.05).④Compared with control group,the protein expressions of PLA2G7,Caspase-1,cleaved-Caspase-3 and p-MLKL in HeLa cells were up-regulated in Melittin group(P<0.05),while these molecules were significantly down-regulated in sh-AIM2 group(P<0.05).Compared with sh-AIM2 group,the low expression of these molecules were reversed in sh-AIM2+Melittin group(P<0.05).Conclusion AIM2 and PLA2G7 are highly expressed and positively correlated in cervical cancer.Down-regulation of AIM2 can reduce PLA2G7 expression,and they synergistically promote PANoptosis in HeLa cells,providing a potential direction for cervical cancer treatment.
