基于高分辨质谱代谢组学及网络药理学的蝉蜕质量评价OA
Quality Evaluation of Cicadae Periostracum Based on High-resolution Mass Spectrometry Metabolomics and Network Pharmacology
蝉蜕为蝉科昆虫黑蚱(Cryptotympana pustulata Fabricius)若虫羽化时脱落的皮壳,药用历史已逾千年.目前,《中国药典》仅以外观性状评价其品质,缺乏与药效关联的规范、科学质控指标.本研究采集8个主产地的蝉蜕样本,经性状与显微鉴定后,采用超高效液相色谱-四极杆飞行时间串联质谱(Ultra Performance Liquid Chromatog-raphy-Quadrupole Time-of-flight Tandem Mass Spectrometry,UPLC-QTQF-MS/MS)代谢组学结合网络药理学方法开展质量评价.结果显示,共鉴定出 52个N-乙酰多巴胺类化合物(含单聚体至五聚体)、29个直链二肽及6个氨基酸;河南开封、四川广安、河北保定的N-乙酰多巴胺类成分含量较高,山西长治、四川广安的特征肽类及氨基酸含量优势显著;网络药理学分析表明,蝉蜕中N-乙酰多巴胺类和二肽类成分作用于原癌基因酪氨酸蛋白激酶(Sarcoma Proto-oncogene Tyrosine-Protein Kinase,SRC)、磷脂酰肌醇3激酶催化亚基α(Phosphatidylinositol-3-Kinase Catalyt-ic Subunit Alpha,PIK3CA)等10个核心靶点,可能通过调控神经活性配体-受体相互作用等通路发挥药理作用,且这两类成分与药理作用关联性较强.综合来看,河南开封、四川广安、河北保定和山西长治四个地区的蝉蜕质量较优,与传统记载"大者入药"一致.本研究建立了高分辨质谱代谢组学与网络药理学相结合的蝉蜕质量评价策略,为其品质评价提供了新的科学依据.
Cicadae Periostracum,the exuviae shed by nymphs of Cryptotympana pustulata Fabricius during eclosion,has been used in traditional medicine for over a thousand years.Currently,its quality is evaluated primarily based on morphological characteristics,lacking standardized and scientifically robust quality control indicators linked to efficacy.In this study,authentic Cicadae Periostra-cum samples from eight major producing areas were collected and identified by morphological and microscopic examination.Ultra-high performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS)was employed for metabolomic profiling,and quality assessment was performed by integrating metabolomics with network pharmacology.A total of 52 N-acetyldopamine compounds(from monomers to pentamers),29 linear dipeptides,and 6 amino acids were identified.Samples from Kaifeng(Henan),Guang'an(Sichuan),and Baoding(Hebei)exhibited relatively higher contents of N-acetyldopamine compounds,while samples from Changzhi(Shanxi)and Guang'an(Sichuan)showed significantly higher levels of characteristic peptides and amino acids.Further network pharmacology analysis revealed that N-acetyldopamine and dipeptide com-ponents act on ten core targets,including Sarcoma Proto-oncogene Tyrosine-Protein Kinase(SRC)and Phosphatidylinositol-3-Ki-nase Catalytic Subunit Alpha(PIK3CA),potentially exerting pharmacological effects through pathways such as neuroactive ligand-receptor interactions.Moreover,these components demonstrated stronger correlations with pharmacological activities.In conclusion,the quality of Cicadae Periostracum from the four regions—Kaifeng(Henan),Guang'an(Sichuan),Baoding(Hebei),and Changzhi(Shanxi)—appears superior,consistent with the traditional notion that"larger specimens are preferable for medicinal use."This study establishes an integrated strategy combining high-resolution mass spectrometry metabolomics and network pharmacology for quality evaluation of Cicadae Periostracum,providing new scientific evidence for its quality assessment.
