目的:探讨蛹虫草提取物调控核因子E2相关因子2(Nrf2)/抗氧化反应元件(ARE)信号通路对老年大鼠骨骼肌缺血再灌注后认知功能的改善作用及机制.方法:将大鼠按照随机数字表法分为假手术组、模型组、蛹虫草提取物低剂量组、蛹虫草提取物高剂量组、蛹虫草提取物高剂量+ML385组,制备肢体缺血再灌注损伤(LIR)模型,用蛹虫草提取物和Nrf2抑制剂ML385分组处理,检测认知功能、腓肠肌组织病理形态、海马神经元凋亡、血清炎症介质水平、腓肠肌与脑组织氧化应激因子水平、腓肠肌与脑组织Nrf2/ARE通路蛋白表达.结果:与假手术组比较,模型组大鼠腓肠肌组织严重损伤,避暗犯错次数、跳台犯错次数、海马神经元凋亡比例、血清-1β(IL-1β)、肿瘤坏死因子α(TNF-α)及IL-6水平、腓肠肌与脑组织丙二醛(MDA)水平高(P<0.05),避暗潜伏期、跳台潜伏期、腓肠肌与脑组织总抗氧化能力(TAC)水平、Nrf2及血红素加氧酶-1(HO-1)蛋白表达低(P<0.05);蛹虫草提取物低、高剂量组大鼠上述病理改变改善,蛹虫草提取物低组作用更强;蛹虫草提取物高剂量+ML385组可消除蛹虫草提取物对LIR模型大鼠各病理指标的改善作用.结论:蛹虫草提取物通过激活Nrf2/ARE信号通路抑制神经炎症与氧化应激,从而减轻老年大鼠骨骼肌缺血再灌注后损伤,并提升其认知功能.

Objective:To investigate the effects and mechanisms of Cordyceps militaris extract in improving cognitive function after skeletal muscle ischemia-reperfusion in elderly rats by regulating the nuclear factor erythroid 2-related factor 2(Nrf2)/antioxidant response element(ARE)signaling pathway.Methods:Rats were randomly divided into a sham operation group,model group,low-dose C.militaris extract group,high-dose C.militaris extract group,and high-dose C.militaris extract+ML385 group using a ran-dom number table.A limb ischemia-reperfusion injury(LIR)model was established.Rats were treated with C.militaris extract and the Nrf2 inhibitor ML385 according to grouping.Cognitive function,pathological morphology of gastrocnemius muscle,apoptosis of hippocampal neurons,serum inflammatory mediator levels,oxidative stress markers in gastrocnemius muscle and brain tissues,and the expression of Nrf2/ARE pathway-related proteins in gastrocnemius muscle and brain tissues were assessed.Results:Compared with the sham operation group,the model group showed severe damage to gastrocnemius muscle tissue.The number of errors in the dark-avoidance and step-down tests,the proportion of hippocampal neuronal apoptosis,serum levels of interleukin-1 β(IL-1 β),tumor necrosis factor-α(TNF-α),and IL-6,as well as malondialdehyde(MDA)levels in gastrocnemius muscle and brain tissues were significantly increased(P<0.05),whereas the latency in the dark-avoidance and step-down tests,total antioxidant capacity(TAC)in gastrocnemius muscle and brain tissues,and the expression of Nrf2 and heme oxygenase-1(HO-1)proteins were signifi-cantly decreased(P<0.05).Both low-and high-dose C.militaris extract improved the above pathological changes,with the low-dose group showing a stronger effect.The high-dose C.militaris extract+ML385 group abolished the protective effects of C.militaris extract on these pathological indicators in LIR model rats.Conclusion:C.militaris extract alleviates skeletal muscle ischemia-reper-fusion injury and improves cognitive function in aged rats by activating the Nrf2/ARE signaling pathway,thereby inhibiting neuroin-flammation and oxidative stress.
