补肾开窍祛痰方治疗缺氧缺血性脑损伤脑瘫的分子机制研究OACHSSCD
Molecular Mechanisms of Bushen Kaiqiao Qutan Formula against Cerebral Palsy Induced by Hypoxic-ischemic Brain Damage
目的:研究补肾开窍祛痰方治疗缺氧缺血性脑损伤(HIBD)脑瘫模型大鼠的分子机制.方法:将大鼠随机分为正常组、假手术组和造模组.采用改良Rice-Vannucci方法制备HIBD脑瘫模型.造模成功的大鼠随机分为模型组、六味地黄汤组及补肾开窍祛痰方低、中、高剂量组.药物干预组术后第2天开始灌胃给药,正常组、假手术组、模型组同时灌胃等量生理盐水,连续干预3周.对比造模后1、2、3周神经功能评分,比较各组海马阿蒙角1(CA1)区Cyt-C、Caspase-9、Case-pase-3、凋亡蛋白酶活化因子-1(Apaf-1)表达.结果:Zea-Longa评分:与模型组比较,正常组、假手术组、补肾开窍祛痰方低、中、高剂量组及六味地黄汤组大鼠第1、2周Zea-Longa评分降低(P<0.05).免疫组化结果:与模型组比较,正常组、假手术组、补肾开窍祛痰方中剂量组大鼠海马CA1区Cyt-C、Caspase-9、Casepase-3、Apaf-1蛋白阳性表达降低.大鼠海马CA1区WB检测结果:与模型组比较,补肾开窍祛痰方中剂量组Cyt-C、Caspase-9、Casepase-3、Apaf-1蛋白表达降低(P<0.01),高剂量组和六味地黄汤组Apaf-1蛋白表达降低(P<0.01).实时荧光定量PCR检测结果:与模型组比较,补肾开窍祛痰方中剂量组大鼠海马CA1区Cyt-C、Caspase-9、Casepase-3、Apaf-1 mRNA表达降低(P<0.01).结论:补肾开窍祛痰方可治疗HIBD脑瘫,可能是通过抑制Cyt-c/Caspase信号通路激活进而抑制细胞凋亡实现的.
Objective:To investigate the molecular mechanisms of Bushen Kaiqiao Qutan Formula(BSKQQTF)in treating a rat model of cerebral palsy induced by hypoxic-ischemic brain damage(HIBD).Methods:Rats were randomly divided into a normal group,a sham operation group,and a modeling group.The HIBD cerebral palsy model was established using the modified Rice-Van-nucci method.Successfully modeled rats were further randomly assigned to a model group,a Liuwei Dihuang Decoction group,and low-,medium-,and high-dose BSKQQTF groups.The drug intervention groups received intragastric administration starting on the second day after surgery,while the normal,sham operation,and model groups were administered an equal volume of normal saline for three consecutive weeks.Neurological function scores were evaluated at 1,2,and 3 weeks after modeling.The expression levels of Cytochrome C(Cyt-C),Caspase-9,Caspase-3,and apoptotic protease activating factor-1(Apaf-1)in the hippocampal CA1 region were assessed in each group.Results:Zea-Longa scores:Compared with the model group,the normal,sham operation,BSKQQTF(low-,medium-,and high-dose),and Liuwei Dihuang Decoction groups showed significantly decreased Zea-Longa scores at weeks 1 and 2(P<0.05).Immunohistochemistry:Compared with the model group,positive expression of Cyt-C,Caspase-9,Caspase-3,and Apaf-1 proteins in the hippocampal CA1 region was reduced in the normal,sham operation,and medium-dose BSKQQTF groups.Western blot for hippocampal CAI region:Compared with the model group,protein expression levels of Cyt-C,Caspase-9,Caspase-3,and Apaf-1 were decreased in the medium-dose BSKQQTF group(P<0.01).In the high-dose BSKQQTF and Liuwei Dihuang Decoction groups,Apaf-1 protein expression was also decreased(P<0.01).Real-time quantitative PCR(qRT-PCR):Compared with the model group,mRNA expression levels of Cyt-C,Caspase-9,Caspase-3,and Apaf-1 in the hippocampal CA1 region were significantly reduced in the medium-dose BSKQQTF group(P<0.01).Conclusion:BSKQQTF can treat HIBD-induced cerebral palsy,possibly by inhibiting neuronal apoptosis through suppression of activation of the Cyt-c/Caspase signaling pathway.
