地黄饮子改善阿尔茨海默病肾虚精亏证大鼠神经炎症的作用机制OACHSSCD
Mechanisms of Dihuang Yinzi Decoction in Improving Neuroinflammation in Rats with Alzheimer's Disease Characterized by Kidney Deficiency and Essence Depletion
目的:探讨地黄饮子对阿尔茨海默病(AD)肾虚精亏证大鼠神经炎症的作用机制.方法:将36只大鼠按照随机数字表法随机分为对照组、AD肾虚精亏证模型组、地黄饮子低剂量组、地黄饮子中剂量组、地黄饮子高剂量组和多奈哌齐组.采用腹腔注射D-半乳糖制备亚急性衰老大鼠结合双侧海马CA1区注射β-淀粉样蛋白片段25-35(Aβ25-35)制备AD肾精虚证病证结合模型.莫里斯(Morris)水迷宫检测大鼠的空间学习记忆能力;免疫荧光检测小胶质细胞活化;免疫组织化学实验检测大鼠海马区β淀粉样多肽1-42(Aβ1-42)和磷酸化微管结合蛋白(Tau)404(pTau404)表达水平;酶联免疫吸附试验(ELISA)检测脑组织中白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平;蛋白质印迹法(WB)检测海马组织中腺苷酸活化蛋白激酶(AMPK)、磷酸化AMPK(p-AMPK)、核因子KBp65(NF-κBp65)、磷酸化NF-κBp65(p-p65)蛋白表达.结果:地黄饮子可显著改善AD肾虚精亏证模型大鼠的空间学习记忆能力;改善海马神经元结构形态,减轻线粒体肿胀;降低离子钙结合适配分子1(Ibal)表达,抑制小胶质细胞活性;减少Aβ1-42和pTau404表达;增加p-AMPK蛋白表达,降低p-p65蛋白表达.结论:地黄饮子可能通过激活AMPK/NF-κB信号通路改善AD肾虚精亏证大鼠神经炎症,从而发挥抗AD作用.
Objective:To investigate the mechanisms of Dihuang Yinzi Decoction on neuroinflammation in rats with Alzheimer's disease(AD)of kidney deficiency and essence depletion syndrome.Methods:Thirty-six rats were randomly divided into a control group,an AD of kidney deficiency and essence depletion syndrome model group,a low-dose Dihuang Yinzi Decoction group,a me-dium-dose Dihuang Yinzi Decoction group,a high-dose Dihuang Yinzi Decoction group,and a donepezil group using a random num-ber table.A combined disease-syndrome model was established by intraperitoneal injection of D-galactose to induce subacute aging in rats,together with bilateral hippocampal CA1 injection of β-amyloid fragment 25-35(Aβ25-3s)to establish an AD model with kid-ney deficiency and essence depletion syndrome.The Morris water maze was used to assess spatial learning and memory ability.Mi-croglial activation was detected by immunofluorescence.The expression levels of β-amyloid 1-42(Aβ1-42)and phosphorylated tau protein 404(pTau404)in the hippocampus were measured by immunohistochemistry.The levels of interleukin-1 β(IL-1 β)and tumor necrosis factor-α(TNF-α)in brain tissue were detected by enzyme-linked immunosorbent assay(ELISA).The protein ex-pression levels of AMP-activated protein kinase(AMPK),phosphorylated AMPK(p-AMPK),nuclear factor-κB p65(NF-κB p65),and phosphorylated NF-κB p65(p-p65)in the hippocampus were detected by Western blot.Results:Dihuang Yinzi Decoction sig-nificantly improved spatial learning and memory ability in AD rats with kidney deficiency and essence depletion syndrome.It im-proved hippocampal neuronal morphology and reduced mitochondrial swelling.The expression of ionized calcium-binding adaptor molecule l(Ibal)was decreased,indicating inhibition of microglial activation.The expression levels of Aβ1-42 and pTau404 were reduced.In addition,p-AMPK protein expression was increased,while p-p65 protein expression was decreased.Conclusion:Di-huang Yinzi Decoction may alleviate neuroinflammation in AD rats with kidney deficiency and essence depletion syndrome by acti-vating the AMPK/NF-κB signaling pathway,thereby exerting anti-AD effects.
