目的:探讨二十味肉豆蔻丸对5个家族性阿尔茨海默病转基因(5xFAD)小鼠学习记忆能力和抑郁样行为的作用.方法:将5xFAD转基因小鼠用简单随机化分为模型组、阳性药组和二十味肉豆蔻丸低、中、高剂量组,野生型(WT)小鼠作为对照组.对照组和模型组小鼠均给予超纯水;阳性药组给予多奈哌齐和美金刚的混悬液;二十味肉豆蔻丸低、中、高剂量组均给予二十味肉豆蔻丸悬浊液,干预334 d.用行为学实验检测学习记忆能力和抑郁样行为.免疫组织化学法检测脑内β-淀粉样蛋白1-42分子量(Aβ1-42)沉积;酶联免疫吸附试验检测促肾上腺激素释放激素(CRH)、促性腺激素释放激素(GnRH)、促甲状腺激素释放激素(TRH)和促甲状腺激素(TSH)以及皮质酮(CORT);放射性免疫分析测定促肾上腺激素(ACTH)、促卵泡激素(FSH)、黄体生成素(LH)和血浆雌二醇(E2)、睾酮(T)、三碘甲状腺原氨酸(T3)和甲状腺素(T4).用Pearson相关法分析指标间的相关性.结果:与模型组比较,二十味肉豆蔻丸组小鼠学习记忆能力、抑郁样行为、脑内Aβ1-42平均光密度均改善(P<0.05),ACTH、FSH、LH、TSH水平降低(P<0.05).相关性结果显示5xFAD小鼠的脑区分泌的激素水平均与学习记忆能力、抑郁样行为及脑区Aβ1-42沉积呈显著相关(|r|>0.5).结论:二十味肉豆蔻丸改善5xFAD小鼠学习记忆能力和抑郁样行为,其作用机制可能与调节神经内分泌功能有关.

Objective:To investigate the effects of Ershiwei Roudoukou Pills on learning and memory abilities and depressive-like behaviors in 5 familial Alzheimer's disease(5xFAD)transgenic mice.Methods:The 5xFAD transgenic mice were randomly divided into a model group,a positive drug group,and low-,medium-,and high-dose Ershiwei Roudoukou Pills groups,with wild-type(WT)mice serving as the control group.Mice in the control and model groups were administered ultrapure water.The positive drug group was administered a suspension of donepezil and memantine.The low-,medium-,and high-dose Ershiwei Roudoukou Pills groups were administered Ershiwei Roudoukou Pills suspensions for an intervention period of 334 days.Behavioral experiments were used to assess learning and memory abilities and depressive-like behaviors.Immunohistochemistry was used to detect amyloid β-protein 1-42(Aβ1-42)deposition in the brain.Enzyme-linked immunosorbent assay(ELISA)was used to measure corticotropin-releasing hormone(CRH),gonadotropin-releasing hormone(GnRH),thyrotropin-releasing hormone(TRH),thyroid-stimulating hormone(TSH),and corticosterone(CORT).Radioimmunoassay was used to determine adrenocorticotropic hormone(ACTH),follicle-stim-ulating hormone(FSH),luteinizing hormone(LH),estradiol(E2),testosterone(T),triiodothyronine(T3),and thyroxine(T4).Pearson correlation analysis was used to evaluate correlations among indicators.Results:Compared with the model group,mice in the Ershiwei Roudoukou Pills groups showed improved learning and memory abilities,alleviated depressive-like behaviors,and re-duced average optical density of Aβ1-42inthe brain(P<0.05),as well as decreased levels of ACTH,FSH,LH,and TSH(P<0.05).Correlation analysis showed that hormone levels in brain regions of 5xFAD mice were significantly correlated with learning and memory abilities,depressive-like behaviors,and Aβ1-42 deposition in these regions(|r|>0.5).Conclusion:Ershiwei Roudouk-ou Pills can improve learning and memory abilities and alleviate depressive-like behaviors in 5xFAD mice,and the mechanism may be related to regulation of neuroendocrine function.
