目的:观察海藻(海蒿子)和甘草(乌拉尔甘草)在2020年版《中华人民共和国药典》最高剂量2倍条件下于海藻玉壶汤中加减应用,对甲状腺肿大大鼠心肾及血脂的影响.方法:将128只维斯塔(Wistar)雄性大鼠按照随机数字表法随机分为空白组、模型组、优甲乐组、海藻玉壶汤(HYD)组、HYD去海藻(HYD-H)组、HYD去甘草(HYD-G)组、HYD去海藻甘草(HYD-HG)组、海藻甘草(HG)组,采用丙硫氧嘧啶(PTU)复制甲状腺肿大病理模型,以优甲乐作为阳性药,其余各中药组给予相应药液灌胃,在末次给药后12 h取材.检测大鼠血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、尿素氮(BUN)、肌酐(Cr)、尿酸(UA)、总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、载脂蛋白A1(ApoA1)、载脂蛋白B(ApoB)水平;苏木精-伊红(HE)染色观察心肾组织病理学变化.结果:与模型组比较,HYD-H组、HYD-HG组、HG组血清BUN水平高(P<0.05,P<0.01),HYD-H组血清Cr水平显著升高(P<0.05),HYD-HG组血清TG、ApoA1水平显著升高(P<0.01),优甲乐组血清ApoA1、ApoB水平显著降低(P<0.05,P<0.01);光镜下各给药组心肾组织病理形态无显著变化.结论:海藻玉壶汤加减海藻甘草反药组合对甲状腺肿大大鼠心肾未产生明显不良反应,对血脂异常无明显改善作用.

Objective:To observe the effects of Sargassum(Sargassum pallidum)and Glycyrrhizae Radix et Rhizoma(Glycyrrhiza uralensis)in Haizao Yuhu Decoction(HYD),administered at twice the maximum dosage specified in the 2020 edition of Chinese Pharmacopoeia,on the heart,kidney,and blood lipids in goiter rats.Methods:A total of 128 male Wistar rats were randomly divid-ed into a blank group,model group,Euthyrox group,HYD group,HYD without Sargassum(HYD-H)group,HYD without Glycyrrhi-zae Radix et Rhizoma(HYD-G)group,HYD without both Sargassum and Glycyrrhizae Radix et Rhizoma(HYD-HG)group,and Sargassum plus Glycyrrhizae Radix et Rhizoma(HG)group.A goiter model was established using propylthiouracil(PTU).Euthyrox was used as the positive control,while the other groups were administered the corresponding decoctions by gavage.Samples were collected 12 h after the final administration.Serum levels of creatine kinase(CK),lactate dehydrogenase(LDH),blood urea nitro-gen(BUN),creatinine(Cr),uric acid(UA),total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),apolipoprotein A1(ApoA1),and apolipoprotein B(ApoB)were measured.His-topathological changes in heart and kidney tissues were observed by hematoxylin-eosin(HE)staining.Results:Compared with the model group,serum BUN levels were increased in the HYD-H,HYD-HG,and HG groups(P<0.05,P<0.01).Serum Cr levels were significantly increased in the HYD-H group(P<0.05).In the HYD-HG group,serum TG and ApoA1 levels were significantly increased(P<0.01).In the Euthyrox group,serum ApoA1 and ApoB levels were significantly decreased(P<0.05,P<0.01).No significant histopathological changes in the heart and kidney tissues were observed in any treatment group under light microscopy.Conclusion:The modified HYD containing the Sargassum-Glycyrrhizae Radix et Rhizoma anti-drug combination did not produce obvious adverse effects on the heart and kidney of goiter rats and showed no significant improvement in dyslipidemia.
