扶阳复圆汤对抑郁模型大鼠肠道菌群及代谢作用的影响OACHSSCD
Effects of Fuyang Fuyuan Decoction on Gut Microbiota and Metabolic Profile in a Rat Model of Depression
目的:基于16S脱氧核糖体DNA(16S rDNA)测序、非靶向代谢组学,探讨扶阳复圆汤抗抑郁的作用机制.方法:将40只斯泼累格·多雷(SD)雄性大鼠随机分为空白组、模型组、氟西汀组以及扶阳复圆汤组.采用孤养结合慢性不可预知温和应激法建立抑郁模型,干预2周后,通过行为学测试(糖水偏好、旷场、悬尾实验)评估抑郁样行为,尼氏染色与苏木精-伊红染色分别观察海马神经元、结肠组织病理变化,通过16S rDNA测序与非靶向代谢组学(LC-MS)技术分析肠道菌群及粪便代谢物,并采用斯皮尔曼相关分析探究菌群与代谢物的关联.结果:与空白组比较,模型组大鼠糖水偏好率、体质量增长及运动距离均显著降低,不动时间延长(P<0.01);扶阳复圆汤干预可明显改善上述行为指标(P<0.05或P<0.01).16S rDNA测序显示,扶阳复圆汤回调厚壁菌门/拟杆菌门比值(F/B),并调节放线菌门和变形菌门丰度.非靶向代谢组学鉴定出137种差异代谢物,主要富集于胆碱代谢和甘油磷脂代谢通路.菌群-代谢物相关性分析表明,黄弯曲菌属、排放物菌科(Defluviitaleaceae)未培养菌群UCG-011等菌属与抑郁相关代谢物显著相关.结论:扶阳复圆汤可通过调节肠道菌群结构、影响F/B比值以及胆碱与甘油磷脂代谢通路改善抑郁样行为,其作用机制与菌群-代谢物互作密切相关.
Objective:To investigate the antidepressant mechanisms of Fuyang Fuyuan Decoction based on 16S ribosomal DNA(16S rDNA)sequencing and non-targeted metabolomics.Methods:Forty male Sprague Dawley(SD)rats were randomly divided into a blank group,a model group,a fluoxetine group,and a Fuyang Fuyuan Decoction group.A depression model was established u-sing solitary housing combined with chronic unpredictable mild stress(CUMS).After 2 weeks of intervention,depressive-like be-haviors were evaluated using behavioral tests,including the sucrose preference test,open field test,and tail suspension test.Nissl staining and hematoxylin-eosin(HE)staining were used to observe pathological changes in hippocampal neurons and colonic tis-sue,respectively.Gut microbiota and fecal metabolites were analyzed using 16S rDNA sequencing and LC-MS-based non-targeted metabolomics.Spearman correlation analysis was performed to explore the relationships between gut microbiota and metabolites.Re-sults:Compared with the blank group,the model group showed significantly decreased sucrose preference,body weight gain,and lo-comotor activity distance,along with prolonged immobility time(P<0.01).Fuyang Fuyuan Decoction intervention significantly im-proved these behavioral indicators(P<0.05 or P<0.01).The 16S rDNA sequencing showed that Fuyang Fuyuan Decoction regu-lated the Firmicutes/Bacteroidetes(F/B)ratio and modulated the abundance of Actinobacteria and Proteobacteria.Non-targeted metabolomics identified 137 differential metabolites,which were mainly enriched in choline metabolism and glycerophospholipid metabolism pathways.Microbiota-metabolite correlation analysis revealed that genera such as Campylobacter,Defluviitaleaceae UCG-011,and other uncultured taxa were significantly associated with depression-related metabolites.Conclusion:Fuyang Fuyuan Decoction can improve depressive-like behaviors by regulating gut microbiota composition,modulating the F/B ratio,and influen-cing choline and glycerophospholipid metabolic pathways.Its antidepressant mechanism is closely associated with microbiota-metab-olite interactions.
