目的:本研究旨在探讨土茯苓(SGR)在从重塑肠道菌群与黏膜免疫稳态方面对溃疡性结肠炎小鼠模型的改善效果和分子机制.方法:通过葡聚糖硫酸钠饲喂小鼠构建溃疡性结肠炎模型,以人体等效剂量预防性灌胃给药土茯苓.通过体质量、疾病活动指数评分、结肠长度以及组织病理学评估肠炎程度,采用阿尔辛蓝/过碘酸希夫染色评估杯状细胞黏膜分泌功能,应用实时荧光定量聚合酶链式反应、流式微球与免疫组织化学检测免疫细胞定量和炎症介质的表达水平,对盲肠内容物进行测序分析.结果:土茯苓显著缓解了小鼠模型的结肠炎症状,保护了杯状细胞功能并恢复了肠道黏液屏障,有效抑制巨噬细胞过度激活,恢复肠炎损伤的伽马-德尔塔T细胞(γδT Cell)与肠上皮细胞的相互作用,降低炎症水平.肠道菌群分析表明,土茯苓通过增加肠道微生态多样性、增加拟杆菌等益生菌群落的丰度、抑制致病性厌氧菌以及调节特定功能菌群来恢复肠炎小鼠模型的肠道微生态失调.结论:土茯苓通过重塑肠道微生态和肠道黏膜免疫,对溃疡性结肠炎小鼠发挥了保护作用,未来有希望用于治疗炎症性肠道疾病.

Objective:To investigate the therapeutic effects and molecular mechanisms of Smilax glabra Roxb.(SGR)in a mouse model of ulcerative colitis(UC),focusing on gut microbiota remodeling and mucosal immune homeostasis.Methods:UC was in-duced in mice by dextran sulfate sodium(DSS),followed by prophylactic oral administration of SGR at a human-equivalent dose.Disease severity was evaluated by body weight,disease activity index(DAI),colon length,and histopathology.Goblet cell function and mucin secretion were assessed using Alcian blue/periodic acid-Schiff(AB/PAS)staining.Immune cell populations and inflam-matory mediator expression were measured by quantitative real-time PCR,multiplex bead-based assays,and immunohistochemistry.Cecal contents were analyzed by 16S rRNA gene sequencing to assess gut microbiota composition.Results:SGR markedly alleviated colonic inflammation,preserved goblet cell function,and restored mucus barrier integrity.It effectively suppressed excessive macro-phage activation,restored γδT cell-epithelial cell interactions disrupted by colitis,and reduced overall inflammatory responses.Gut microbiota analysis revealed that SGR increased microbial diversity,enhanced the abundance of beneficial taxa such as Bacte-roides,suppressed pathogenic anaerobes,and modulated specific functional bacterial communities,thereby correcting dysbiosis in UC mice.Conclusion:SGR exerts protective effects against UC by reshaping gut microbiota and restoring mucosal immune homeo-stasis,indicating its potential as a therapeutic candidate for inflammatory bowel disease.
