目的:探讨芹菜籽和槐米提取物(芹槐提取物)在治疗酒精性肝病(ALD)和代谢功能障碍相关脂肪性肝病(MASLD)中的作用.方法:采用苏木精-伊红(HE)染色和油红染色观察芹槐提取物对Gao-Binge小鼠ALD模型和高脂饲料诱导的小鼠MASLD模型中肝内脂肪变性和肝脏病理变化的影响.通过检测血清谷丙转氨酶(GPT)、谷草转氨酶(GOT)以及三酰甘油等指标,评价芹槐提取物对肝脂肪变性和肝损伤的改善效果.结果:在ALD模型中,芹槐提取物显著缓解了乙醇摄入导致的以大泡为主的混合型脂肪变性、局灶性气球样变、细胞核固缩以及肝索结构紊乱等病理改变,并显著降低血清GOT水平,对GPT和血清三酰甘油也表现出一定的改善作用.在MASLD模型中,芹槐提取物显著减轻了高脂饮食诱导的混合型脂肪变性,并有效缓解了广泛存在的肝细胞气球样变、细胞核固缩以及肝索结构异常等病变,且呈现剂量依赖性.在生化指标上,芹槐提取物显著降低了血清GPT和GOT水平,同时降低了血清三酰甘油和总胆固醇等脂肪变性相关指标.结论:芹槐提取物在小鼠ALD和MASLD模型中表现出显著的治疗作用,能够有效减轻肝内脂肪变性并缓解肝损伤.这一结果与既往芹槐提取物相关临床试验相一致,表明该提取物除治疗痛风和高尿酸血症外,还具有良好的防治脂肪性肝病的潜力.

Objective:To investigate the effects of Qinhuai(QH,Apium graveolens seed and Sophora japonica bud)extract on alco-holic liver disease(ALD)and metabolic dysfunction-associated steatotic liver disease(MASLD).Methods:Hematoxylin and eosin(HE)staining and Oil Red O staining were used to evaluate the effects of QH on hepatic steatosis and pathological changes in the Gao-Binge ALD mouse model and high-fat diet(HFD)-induced MASLD mouse model.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),and triglycerides(TG)were measured to assess the ameliorative effects of QH on liver steatosis and liver injury.Results:In the ALD model,QH significantly alleviated alcohol-induced pathological changes,including mixed-type steatosis predominantly characterized by macrovesicular fat droplets,focal hepatocyte ballooning,nuclear pyknosis,and disrupted hepatic cords.It significantly reduced serum AST levels and moderately improved ALT and TG levels.In the MASLD model,QH markedly reduced HFD-induced mixed-type steatosis and effectively mitigated widespread hepatocyte ballooning,nuclear pyknosis,and disrupted hepatic cord structures in a dose-dependent manner.Biochemically,QH significantly lowered serum ALT and AST levels and reduced steatosis-related indicators,including TG and total cholesterol(T-CHO).Conclusion:QH extract dem-onstrates significant therapeutic effects in mouse models of ALD and MASLD by effectively reducing hepatic steatosis and allevia-ting liver injury.These results are consistent with previous clinical studies and suggest that,in addition to its established roles in treating gout and hyperuricemia,QH extract may also have potential for the prevention and management of fatty liver disease.
