目的:探讨桂枝茯苓汤调节血管内皮生长因子/磷脂酰肌醇3-激酶/蛋白激酶B(VEGF/PI3K/AKT)信号通路对精索静脉曲张(VC)模型大鼠生精细胞凋亡的影响.方法:斯泼累格·多雷(SD)大鼠随机分为5组,包括正常大鼠组(NC组)、VC模型组(VC组)、VC+桂枝茯苓汤低剂量组(GZFL-L组)、VC+桂枝茯苓汤中剂量组(GZFL-M组)和VC+桂枝茯苓汤高剂量组(GZFL-H组),全自动生化检测仪检测睾丸激素水平,TUNEL试剂盒检测大鼠睾丸生精细胞凋亡,免疫荧光或免疫组织化学方法检测VEGF、AKT、磷酸化AKT(p-AKT)、p70核糖体S6蛋白激酶(p70S6K)和磷酸化p70S6K(p-p70S6K)的表达.结果:与NC组比较,VC组的大鼠睾丸激素水平显著降低,经过不同剂量的GZFL处理后,GZFL-L组、GZFL-M组和GZFL-H组的大鼠睾丸激素水平均较VC组有所提高,其中GZFL-H组的提高最为显著(P<0.05).与NC组比较,VC组生精细胞凋亡数明显增加(P<0.05).而与VC组比较,GZFL-L组、GZFL-M组和GZFL-H组凋亡细胞减少(P<0.05).与NC组比较,VC组VEGF蛋白表达增加;GZFL-L组、GZFL-M组和GZFL-H组VEGF的表达明显降低(P<0.05).此外,与NC组比较,VC组p-AKT和p-p70S6K的表达也增加(均P<0.05).与VC组比较,GZFL-L组、GZFL-M组和GZFL-H组p-AKT和p-p70S6K蛋白表达下降(P<0.05).结论:桂枝茯苓汤可能通过阻断VEGF/PI3K/AKT信号通路抑制VC大鼠生精细胞凋亡.

Objective:To investigate the effects of Guizhi Fuling Decoction on spermatogenic cell apoptosis in a rat model of vari-cocele(VC)via regulation of the vascular endothelial growth factor/phosphatidylinositol 3-kinase/protein kinase B(VEGF/PI3K/AKT)signaling pathway.Methods:Sprague Dawley(SD)rats were randomly divided into five groups:normal control group(NC),VC model group(VC),VC+Guizhi Fuling Decoction low-dose group(GZFL-L),VC+Guizhi Fuling Decoction medium-dose group(GZFL-M),and VC+Guizhi Fuling Decoction high-dose group(GZFL-H).Testicular testosterone levels were measured using an automatic biochemical analyzer.Apoptosis of spermatogenic cells in rat testes was detected using a TUNEL assay.The expression levels of VEGF,AKT,phosphorylated AKT(p-AKT),p70 ribosomal S6 kinase(p70S6K),and phosphorylated p70S6K(p-p70S6K)were determined by immunofluorescence or immunohistochemistry.Results:Compared with the NC group,testicular testosterone levels in the VC group were significantly decreased.After treatment with different doses of Guizhi Fuling Decoction,testosterone lev-els in the GZFL-L,GZFL-M,and GZFL-H groups were increased compared with those in the VC group,with the most significant in-crease observed in the GZFL-H group(P<0.05).Compared with the NC group,the number of apoptotic spermatogenic cells was significantly increased in the VC group(P<0.05).Compared with the VC group,the number of apoptotic cells in the GZFL-L,GZ-FL-M,and GZFL-H groups was significantly reduced(P<0.05).Compared with the NC group,VEGF protein expression was in-creased in the VC group,whereas VEGF expression was significantly decreased in the GZFL-L,GZFL-M,and GZFL-H groups(P<0.05).In addition,compared with the NC group,the expression levels of p-AKT and p-p70S6K were increased in the VC group(both P<0.05).Compared with the VC group,the expression levels of p-AKT and p-p70S6K were decreased in the GZFL-L,GZ-FL-M,and GZFL-H groups(P<0.05).Conclusion:Guizhi Fuling Decoction may inhibit spermatogenic cell apoptosis in VC rats by blocking the VEGF/PI3K/AKT signaling pathway.
