不同制备工艺苓桂术甘汤化学成分分析与方剂配伍合理性分析OACHSSCD
Chemical Composition of Linggui Zhugan Decoction with Different Preparation Processes and Rationality of Its Formula Compatibility
目的:比较不同制备工艺下苓桂术甘汤物质基础的相似性与差异性,并基于网络药理学方法阐明物质基础分布与配伍的潜在联系.方法:建立苓桂术甘汤超高效液相色谱-四极杆-飞行时间串联质谱法(UHPLC-QTOF-MS/MS)靶向测定方法,对苓桂术甘汤传统基准样品与智能代煎样品进行定性分析和相对定量分析,并基于SWISS等网络药理学工作站筛选关键活性成分和对非酒精性脂肪肝(NAFLD)关键基因进行基因本体(GO)富集分析.结果:苓桂术甘汤传统基准样品与智能代煎样品均各自含有142种成分,共性成分占91.89%,化学轮廓高度相似.相对定量结果则表明智能代煎工艺改善了传统水煎工艺中茯苓成分低溶出的问题.此外,GO富集分析显示茯苓生物学过程中所涉及基因数最多,介数中心度最高,提示茯苓"药力居首",其他药味则对其产生协同与辅助作用.结论:基于UHPLC-QTOF-MS/MS新技术路线,从定性成分-相对定量系统分析了苓桂术甘汤不同制备工艺下物质基础的相似性与差异性.基于网络药理学,从物质基础分布的角度阐明了苓桂术甘汤对NAFLD组方配伍的合理性,可作为其他中药汤剂化学成分研究之参考.
Objective:To compare the similarities and differences in the material basis of Linggui Zhugan Decoction(LGZG)pre-pared using different methods,and to clarify the potential relationship between material distribution and compatibility based on net-work pharmacology.Methods:A targeted determination method for LGZG was established using ultra-high-performance liquid chro-matography coupled with quadrupole time-of-flight tandem mass spectrometry(UHPLC-QTOF-MS/MS).Qualitative and relative quantitative analyses were conducted on traditional benchmark samples and intelligent decoction samples.Key active ingredients were screened,and Gene Ontology(GO)enrichment analysis of targets related to non-alcoholic fatty liver disease(NAFLD)was performed using network pharmacology platforms,including SWISS.Results:Both the traditional benchmark and intelligent decoc-tion samples contained 142 components,with 91.89%overlap,indicating highly similar chemical profiles.Relative quantitative a-nalysis showed that the intelligent decoction process improved the dissolution of Poria cocos components,which were poorly soluble in the traditional water decoction.GO enrichment analysis revealed that P.cocos was associated with the largest number of genes and exhibited the highest betweenness centrality,suggesting it plays the principal therapeutic role,while the other herbs provide synergistic and auxiliary effects.Conclusion:Using UHPLC-QTOF-MS/MS,this study systematically analyzed the similarities and differences in the material basis of LGZG prepared by different methods at both qualitative and relative quantitative levels.Network pharmacology analysis clarified the rationale for LGZG compatibility in treating NAFLD from the perspective of material distribu-tion,providing a reference for chemical composition studies of other traditional Chinese medicine decoctions.
