肺腺癌组织中CCT7表达变化的预测与临床验证OA
Prediction and clinical validation of CCT7 expression changes in lung adenocarcinoma tissues
目的 基于生物信息学数据库预测肺腺癌组织中含有 T-复合物多肽 1 的伴侣蛋白 7(CCT7)的表达变化,并进行临床验证.方法 通过 GDC 数据库下载 TCGA 肺腺癌项目的 58 例肺腺癌患者的转录组测序数据,比较肺腺癌组织及癌旁组织中 CCT7 mRNA 的表达变化.从 UCSC Xena 数据库下载 TCGA 肺腺癌患者的临床病理分期信息,并与 CCT7 mRNA 表达谱数据进行整合,分析 CCT7 mRNA 表达与肺腺癌患者临床病理分期的关系.基于TCGA 数据库 CCT7 mRNA 表达谱数据,以中位表达量为截断值,将肺腺癌患者分为 CCT7 高表达组和低表达组,分析 CCT7 mRNA 表达与肺腺癌患者预后的关系.收集 2023 年 9 月—2025 年 8 月徐州医科大学附属徐州市立医院收治的 30 例肺腺癌患者的癌组织及配对癌旁组织样本,采用实时荧光定量 PCR 法检测肺腺癌组织中 CCT7 mRNA,采用Western blotting 法检测肺腺癌组织中CCT7 蛋白,并分析CCT7 mRNA 和蛋白相对表达量与肺腺癌患者临床病理分期的关系.结果 TCGA 数据库分析结果显示,肺腺癌组织中 CCT7 mRNA 相对表达量高于癌旁组织(P<0.05),Ⅲ期和Ⅳ期肺腺癌组织中 CCT7 mRNA 相对表达量均高于Ⅰ期(P 均<0.05),CCT7 高表达组中位总生存期低于 CCT7 低表达组(P<0.05).30 例肺腺癌患者的癌组织中 CCT7 mRNA 和蛋白相对表达量均高于癌旁组织(P 均<0.05),Ⅰ~Ⅱ期肺腺癌组织中 CCT7 mRNA 和蛋白相对表达量低于Ⅲ~Ⅳ期(P<0.05).结论 CCT7 在肺腺癌组织中高表达,其表达水平与肺腺癌患者的临床病理分期及预后有关.
Objective Based on bioinformatics databases,we predicted the expression changes of chaperonin contai-ning T-complex polypeptide 1 subunit 7(CCT7)in lung adenocarcinoma tissues,and subsequently performed clinical vali-dation.Methods The transcriptome sequencing data of 58 patients with lung adenocarcinoma from the TCGA lung adeno-carcinoma project were downloaded from the GDC database,and the expression changes of CCT7 mRNA in lung adenocarci-noma tissues and adjacent non-cancerous tissues were compared.Clinicopathological staging information of TCGA lung ade-nocarcinoma patients was downloaded from the UCSC Xena database and integrated with the CCT7 mRNA expression profile data to analyze the relationship between CCT7 mRNA expression and clinicopathological staging of lung adenocarcinoma pa-tients.Based on the CCT7 mRNA expression profile data from the TCGA database,lung adenocarcinoma patients were di-vided into the high CCT7 expression group and low CCT7 expression group using the median expression level as the cutoff value,and the relationship between CCT7 mRNA expression and prognosis of lung adenocarcinoma patients was analyzed.Cancer tissue and paired adjacent non-cancerous tissue samples were collected from 30 patients with lung adenocarcinoma who were admitted to The Affiliated Xuzhou Municipal Hospital of Xuzhou Medical University from September 2023 to Au-gust 2025.Real-time quantitative PCR was used to detect CCT7 mRNA in lung adenocarcinoma tissues,and Western blot-ting was used to detect CCT7 protein in lung adenocarcinoma tissues.The relationships between the relative expression lev-els of CCT7 mRNA and protein and the clinicopathological staging of lung adenocarcinoma patients were also analyzed.Results Analysis of the TCGA database showed that the relative expression level of CCT7 mRNA in the lung adenocarcino-ma tissues was higher than that in the adjacent non-cancerous tissues(P<0.05);the relative expression levels of CCT7 mRNA in stage Ⅲ and stage Ⅳ lung adenocarcinoma tissues were both higher than that in stage Ⅰ(both P<0.05).The median overall survival of the high CCT7 expression group was lower than that of the low CCT7 expression group(P<0.05).In the 30 patients with lung adenocarcinoma,the relative expression levels of CCT7 mRNA and protein in the cancer tissues were higher than those in the adjacent non-cancerous tissues(both P<0.05).The relative expression levels of CCT7 mRNA and protein in stage Ⅰ-Ⅱ lung adenocarcinoma tissues were lower than those in stage Ⅲ-Ⅳ(all P<0.05).Conclusion CCT7 is highly expressed in lung adenocarcinoma tissues,and its expression level is associated with the clinicopathological stage and prognosis of patients with lung adenocarcinoma.
