目的 基于生物信息学数据库观察肺腺癌(LUAD)组织中 TANC2 的表达变化,并探讨其意义.方法 使用UALCAN 数据库,选定The Cancer Genome Atlas(TCGA)数据集,分析TANC2 mRNA 在LUAD 与正常肺组织中的表达变化.使用人类蛋白质图谱(HPA)数据库,分析 TANC2 蛋白在 LUAD 与正常肺组织中的归一化蛋白表达量(nRPX).基于 UALCAN 数据库,分析 TANC2 mRNA 相对表达量与 LUAD 患者临床病理参数的关系.基于 HPA数据库,分析 TANC2 mRNA 相对表达量与 LUAD 患者预后的关系.使用 LinkedOmics 数据库,筛选 TANC2 基因在LUAD 中的共表达基因,并进行 KEGG 信号通路富集分析.结果 LUAD 组织中 TANC2 mRNA 相对表达量、TANC2蛋白 nRPX 值均高于正常肺组织(P 均<0.001).女性 LUAD 患者肿瘤组织中 TANC2 mRNA 相对表达量显著高于男性患者,Ⅲ期 LUAD 患者肿瘤组织中 TANC2 mRNA 相对表达量显著高于Ⅳ期患者(P 均<0.05).TANC2 mRNA高表达组患者 5 年生存率低于低表达组(P<0.05).共表达基因筛选结果显示,5 219 个基因与 TANC2 基因表达呈显著正相关(P 均<0.05),5 419 个基因与 TANC2 基因表达呈显著负相关(P 均<0.05),其中 DDX42 基因与 TANC2基因的正相关性最强(r=0.611 5,P<0.05),COMMD7 基因与 TANC2 基因的负相关性最强(r=-0.545 0,P<0.05).KEGG 信号通路富集分析结果显示,TANC2 可能与癌症相关微小 RNA 调控通路、黏附连接通路、甲状腺激素信号通路等多条信号通路的活化有关.结论 TANC2 在 LUAD 组织中高表达,其高表达与 LUAD 患者不良预后有关.TANC2 在LUAD 组织中的表达变化与DDX42、COMMD7 等基因有关,可能参与了微小RNA 等癌症相关信号通路的调控.

Objective To observe the expression changes of TANC2 in lung adenocarcinoma(LUAD)tissues based on bioinformatics databases and to investigate its clinical significance.Methods The expression changes of TANC2 mRNA in LUAD and normal lung tissues were analyzed using the UALCAN database,based on The Cancer Genome Atlas(TC-GA)dataset.The normalized relative protein expression(nRPX)levels of TANC2 protein in LUAD and normal lung tissues were analyzed using the Human Protein Atlas(HPA)database.The relationship between TANC2 mRNA expression and clinicopathological parameters of LUAD patients was analyzed using the UALCAN database.Based on the HPA database,the correlation between the relative expression level of TANC2 mRNA and the prognosis of patients with LUAD was ana-lyzed.Co-expressed genes of TANC2 in LUAD were identified using the LinkedOmics database,followed by KEGG signa-ling pathway enrichment analysis.Results Both TANC2 mRNA expression and TANC2 protein nRPX levels were signifi-cantly higher in the LUAD tissues than in the normal lung tissues(both P<0.001).TANC2 mRNA expression was signifi-cantly higher in female LUAD patients than in male patients,and was also significantly higher in stage III patients than in stage IV patients(all P<0.05).The 5-year survival rate of patients in the high TANC2 mRNA expression group was signifi-cantly lower than that in the low expression group(P<0.05).Co-expression analysis revealed that 5,219 genes were signifi-cantly positively correlated with TANC2 expression(P<0.05),among which DDX42 exhibited the strongest correlation(R=0.611 5).Conversely,5,419 genes were significantly negatively correlated with TANC2(P<0.05),and COMMD7 exhibited the strongest negative correlation(R=-0.545 0).KEGG pathway enrichment analysis revealed that TANC2 might be involved in the activation of multiple signaling pathways,including cancer-related microRNA regulation pathway,adher-ens junction pathway,and thyroid hormone signaling pathway.Conclusions TANC2 is highly expressed in LUAD tissues,and its elevated expression is associated with poor prognosis in LUAD patients.In addition,the expression changes of TANC2 in LUAD are related to DDX42 and COMMD7 genes,and may participate in the regulation of cancer-related signa-ling pathways,such as the microRNA pathway in cancer.
