常规授粉下不同植物激素处理对烟草种间杂交花粉管生长和结实率的影响OA
Effects of Phytohormone Application during Conventional Pollination on Pollen Tube Growth and Seed Setting Rate in Nicotiana Interspecific Hybridization
种间杂交育种是拓宽遗传资源多样性的有效手段.为了寻找提高烟草种间杂交结实率的方法,本试验设置常规授粉及常规授粉前后单独或组合施用植物激素GA3、IAA、6-BA共4个处理,研究其对Nicoti-ana tabacum cv.K326(母本)与 N.rustica var.brasilia、N.glutinosa、N.longiflora(父本)种间杂交结实率的影响.结果显示,T3处理(250 mg/L GA3涂抹柱头+常规授粉+24 h后用200 mg/L IAA喷施柱头+72 h后用20 mg/L 6-BA喷施柱头)能显著提高3个杂交组合的结实率.以K326为母本,以N.glutinosa和N.rustica var.brasilia为父本种间杂交的花粉管荧光观察结果显示,授予父本花粉后4~12 h间花粉活力受到抑制,24 h花粉活力恢复,48 h花粉管萌发,72 h花粉管进入花柱组织;父本花粉与母本柱头有不亲和现象,花粉管在花柱的1/4处停止生长,并产生大量分枝,花粉管尖端出现膨大、弯曲、胼胝质塞现象.与对照(常规授粉)相比,T1处理(250 mg/L GA3涂抹柱头+常规授粉)能够促进花粉萌发,但对花粉管生长影响不明显;相比于T1处理,T2处理(250 mg/L GA3涂抹柱头+常规授粉+24 h后用200 mg/L IAA喷施柱头)对花粉管生长影响也不明显;相比于T2处理,T3处理能促进花粉管在雌蕊中的生长.本研究结果可为今后提高烟草种间杂交结实率和烟草品质改良提供一定的理论依据.
Interspecific hybridization is an effective means to broaden the diversity of genetic resources in Nicotiana.To find methods for improving seed setting rate of interspecific hybridization,four treatments were established in this study including conventional pollination(CK)and individual or combined application of plant hormones GA3,IAA and 6-BA before and after conventional pollination.The effects of these treatments on seed setting rate were investigated in the crosses between N.tabacum cv.K326(as the female parent)and N.rustica var.brasilia,N.glutinosa,or N.longiflora(as the male parents).The results showed that the T3 treatment(250 mg/L GA3 applied to the stigma+conventional pollination+200 mg/L IAA sprayed onto the stigma at 24 h of pollination+20 mg/L 6-BA sprayed onto the stigma at 72 h of pollination)significantly in-creased the seed setting rate in all the three hybrid combinations.Fluorescence microscopy of pollen tubes in the crosses with K326 as the female parent and N.glutinosa or N.rustica var.brasilia as the male parents showed that the pollen viability was inhibited during 4 to 12 h after pollination but recovered at 24 h,and the pollen tube germinated at 48 h and entered the stylar tissues at 72 h.The incompatibility reaction was observed between the paternal pollen and the maternal stigma:the pollen tubes stopped growing upon reaching about one-quarter of the style length,numerous branches produced,and their tips exhibited swelling,bending and callose plug deposition.Compared with the control(CK),the T1 treatment(250 mg/L GA3 applied to the stigma+conventional pollination)promoted pollen germination but had a negligible effect on pollen tube growth.Compared with T1,the T2 treatment(250 mg/L GA3 applied to the stigma+conventional pollination+200 mg/L IAA sprayed onto the stigma at 24 h of pollination)showed no significant improvement in pollen tube growth.In contrast,the T3 treatment markedly enhanced pollen tube growth within the pistils compared with the T2 treatment.The results of this study could provide a theoretical foundation for improving the seed setting rate of Nicotiana interspecific hybridization and improving the tobacco quality in future.
