章丘大葱地方品种'大梧桐'染色体多样性分析OA

染色体变异是作物进化和育种选择的重要遗传基础.地方品种'大梧桐'是国家地理标志产品章丘大葱的核心种质,具有重要的经济价值,但关于其染色体组成及多样性的研究鲜有报道.本研究采用基因组测序结合染色体荧光原位杂交(FISH)技术,对其染色体特征进行系统解析.结果表明,从'大梧桐'基因组中共鉴定出288个串联重复序列簇,成功开发得到15条特异性寡核苷酸探针;组配了包含9条探针的寡核苷酸探针套,构建了'大梧桐'的细胞学参考核型.对33个供试单株的染色体多态性分析结果表明,'大梧桐'不同个体间存在丰富的核型变异,染色体Chr1—Chr4未检测到核型多态性,其余4条染色体共鉴定出14种多态类型.其中Chr8的多态类型最为丰富(6种),其次为Chr7(4种),Chr5和Chr6各含2种类型.3种多态类型(Chr5-1、Chr 6-1和Chr 8-1)在群体中的频率超过50%,可作为'大梧桐'品种群体的优势类型.供试单株染色体平均纯合率、杂合率和异质率分别为67.51%、32.49%和15.91%;Chr7的杂合率最高(66.67%),Chr8的异质率最高(27.27%),表明该品种具有较高的遗传多样性.本研究结果可为章丘大葱种质资源鉴定、遗传研究及改良提供重要的技术支撑.

Chromosome variation is one of the important genetic factors for crop evolution and breeding selection.The landrace Dawutong is a core germplasm of the national geographical indication cultivar Zhangqiu green onion(Allium fistulosum L.)with significant economic value.However,its chromosome constitution and diversity remain unclear.In this study,its chromosome characteristics were systematically analyzed using ge-nome sequencing and fluorescence in situ hybridization(FISH).The results were as follows.A total of 288 tandem repeat sequence clusters were identified and 15 oligonucleotide probes were designed.According to the signal patterns of these probes,an oligonucleotide probe multiplex containing nine probes were developed for distinguishing chromosomes of Dawutong,and a reference karyotype was constructed.Karyotyping analysis of 33 individual plants revealed that Dawutong had a high degree of karyotype variation among different individu-als.Chr1 to Chr4 each showed one karyotype,while the other four chromosomes contained 14 karyotypes.Chr8 had the most karyotypes(6),followed by Chr7(4),and Chr5 and Chr6 each contained 2 karyotypes.Three polymorphic types(Chr5-1,Chr6-1 and Chr8-1)had the frequency more than 50%,representing the domi-nant ones in Dawutong population.The average rates of chromosome homozygosity,heterozygosity and hetero-geneity were 67.51%,32.49%and 15.91%,respectively.Chr7 had the highest heterozygosity rate as 66.67%,and Chr8 had the highest heterogeneity rate as 27.27%,indicating high genetic diversity of this landrace.These results could provide an important technical support for Zhangqiu green onion germplasm identification,genetic research and improvement.
