邵阳鸡源奇异变形杆菌分离鉴定及生物学特性研究OA
Isolation,Identification and Biological Characteristics of Proteus mirabilis from Poultry in Shaoyang
本研究以湖南省邵阳市地区鸡源奇异变形杆菌(Proteus mirabilis,PM)为研究对象,阐明其流行特征、耐药表型、生物被膜形成能力及毒力特征,为该菌在家禽养殖中的精准防控与抗菌药物合理应用提供科学依据.试验选取邵阳地区生态条件相近的 6 个规模化蛋鸡养殖场(75 日龄),采集肛拭子样品 153 份,经分离培养、生化鉴定及 16S rDNA测序鉴定后,对分离菌株系统开展生物被膜形成能力测定、药物敏感性试验及耐药基因与毒力基因检测.结果显示,邵阳地区鸡源奇异变形杆菌分离率达 37.9%(58/153),多数菌株具有典型迁徙生长表型,其中强生物被膜形成菌株占比 50.0%;药敏试验表明,分离株耐药现象显著,仅对美罗培南耐药率相对较低(24.14%),对磺胺异噁唑耐药率高达 100.00%;多重耐药现象严重,89.7%(52/58)菌株对 8 种及以上抗菌药物耐药;基因检测共检出 84种耐药基因及 56 种毒力与定植相关基因.综上,邵阳地区鸡源奇异变形杆菌已呈高耐药、高毒力复合表型,提示需加强区域性监测并制定精准防控策略,减少其对养鸡产业的危害及潜在公共卫生安全风险.
This study focuses on chicken-derived Proteus mirabilis(PM)in Shaoyang,Hunan,aiming to clarify its epidemiological characteristics,drug-resistant phenotypes,biofilm-forming abilities,and virulence characteristics,thereby providing scientific support for the precise prevention and control of this bacterium in poultry farms and the rational use of antibacterial agents.For the experiment,6 large-scale farms with 75-day-old layer hens in Shaoyang,which had basically consistent ecological conditions,were selected,and 153 anal swab samples were collected.After isolation and culture,biochemical identification,and 16S rDNA sequencing verification to obtain the target strains,systematic tests were conducted on the isolated strains,including determination of biofilm-forming ability,drug sensitivity test,and detec-tion of drug-resistant genes and virulence genes,so as to complete the strain screening and characteristic analysis through-out the process.The results showed that the isolation rate of chicken-derived Proteus mirabilis in Shaoyang was 37.9%(58/153).Most strains exhibited a typical swarming growth phenotype,among which strains with strong biofilm-forming ability accounted for a prominent proportion,reaching 50.0%.The drug sensitivity test indicated that the isolated strains had significant drug-resistant characteristics:the drug resistance rate to meropenem was relatively low(24.14%),while the drug resistance rate to sulfafurazole was as high as 100.00%.The multi-drug resistance phenomenon was serious,with 89.7%(52/58)of the strains being resistant to 8 or more antibiotics.Gene detection revealed that the 58 strains carried a total of 84 types of drug-resistant genes and 56 types of virulence and colonization-related genes.This study indicates that chicken-derived Proteus mirabilis in Shaoyang has shown a composite phenotype of high drug resistance and high virulence.It is urgent to strengthen regional monitoring and explore new precise prevention and control strategies to re-duce its harm to the poultry industry and potential public health risks.
