黑色素生成抑制肽的设计筛选及功效评价研究OA
Design,screening and efficacy evaluation of melanin-production inhibitory peptides
针对目前具有黑色素抑制活性四肽的相关研究较少,文章以黑色素抑制活性为研究目标,通过构建虚拟肽库、建立设计筛选方法,对具有黑色素抑制活性的四肽进行筛选及功效评价.文章以虚拟肽库为基础,借助分子对接、活性预测、作用力分析等技术手段,对目标活性四肽进行筛选,再利用固相肽合成法对目标四肽进行定向合成,进一步从生化检测、细胞模型、分子转录等角度,对目标活性四肽进行功效评价.经筛选获得4条四肽:肽-1(CHWH)、肽-2(RVGW)、肽-3(LHSY)和肽-4(LFSY).相比其他三条四肽,肽-1表现出更好的Cu2+螯合、ABTS阳离子自由基清除和酪氨酸酶抑制活性.同时,其在B16细胞模型内的酪氨酸酶活性和黑色素生成抑制的IC50分别为0.28和0.20g/L,且在10 g/L质量浓度以下对细胞增殖率无显著影响.转录水平研究也进一步显示,肽-1能够降低细胞内MITF(小眼畸形转录因子)和TYR(酪氨酸酶)基因表达水平,且呈浓度依赖性.
Currently,there is little research on tetrapeptides with melanin inhibitory activity.This article aims to study melanin inhibition by constructing a virtual peptide library and establishing a design and screening method to select and evaluate tetrapeptides with melanin inhibitory activity.Based on a virtual peptide library,technologies such as molecular docking,activity prediction,and force analysis were used to screen the target active tetrapeptides,and then solid-phase peptide synthesis was employed for directed synthesis of the target tetrapeptides.The efficacy of the target active tetrapeptides was further evaluated from biochemical detection,cell models,and molecular transcription perspectives.Four tetrapeptides are selected:Peptide-1(CHWH),Peptide-2(RVGW),Peptide-3(LHSY),and Peptide-4(LFSY).Compared to the other three tetrapeptides,Peptide-1 exhibits better Cu2+chelation,ABTS positive ion scavenging,and tyrosinase inhibitory activity.The IC50 values for tyrosinase activity and melanin production inhibition in B16 cell models are 0.28 and 0.20 g/L,respectively,and it dose not significantly affect cell proliferation below a mass concentration of 10 g/L.Transcriptional studies also further show that Peptide-1 can decrease the intracellular levels of MITF(microphthalmia-associated transcription factor)and TYR(tyrosinase)gene expression in a concentration ration-dependent manner.
