OGG1调控心肌细胞增殖的功能研究OA
Functional Research on the Regulation of Cardiomyocyte Proliferation by OGG1
为探究8-氧鸟嘌呤DNA糖基化酶1(8-Oxoguanine DNA glycosylase 1,OGG1)对心肌细胞增殖的调控功能,通过转染si-OGG1小干扰 RNA(OGG1-si)和 OGG1过表达腺病毒(OGG1-OE),分别构建 OGG1敲低和过表达的原代心肌细胞模型.采用实时定量PCR(qPCR)技术检测敲低和过表达的效率,并通过增殖标志物(Ki67和pH3)的表达水平评估原代心肌细胞增殖状态.研究结果显示,与对照组相比,OGG1敲低组中增殖标志物 Ki67和pH3的表达显著降低,而 OGG1过表达组中二者表达均显著升高,表明 OGG1对心肌细胞增殖具有关键调控作用,其过表达能够显著促进心肌细胞的增殖,敲低则抑制心肌细胞的增殖.
To investigate the regulatory function of 8-oxoguanine DNA glycosylase 1(OGG1)on cardiomyocyte proliferation,si-OGG1 small interfering RNA(OGG1-si)and OGG1 overexpression adenovirus(OGG1-OE)were transfected to construct primary car-diomyocyte models with OGG1 knockdown and overexpression,respectively.The effi-ciency of knockdown and overexpression was detected by real-time quantitative PCR(qPCR),and the proliferation status of primary cardiomyocytes was evaluated by the ex-pression levels of proliferation markers(Ki67 and pH3).The results show that compared with the control group,the expression of proliferation markers Ki67 and pH3 is signifi-cantly decreased in the OGG1 knockdown group,while both of them significantly increase in the OGG1 overexpression group.This indicates that OGG1 has a crucial regulatory role in cardiomyocyte proliferation,and its overexpression can significantly promote cardio-myocyte proliferation,while knockdown inhibits cardiomyocyte proliferation.
