小麦耐低温相关NAC转录因子分析与表达OA
Analysis and Expression of NAC Transcription Factors Related to Low Temperature Tolerance in Wheat
为探究小麦耐低温相关NAC转录因子的功能特性,选用耐低温品种蚂蚱麦和低温敏感品种济麦22进行转录组测序,筛选出31个可能与耐低温相关的NAC转录因子基因家族成员.生物信息学分析显示,31个TaNAC蛋白中12个为碱性蛋白(pI>7),其余均为酸性蛋白(pI<7);所有蛋白均呈亲水性,主要定位于细胞核,二级结构以无规则卷曲为主.共线性分析表明,小麦与水稻的NAC转录因子基因之间具有较多的共线性关系.启动子分析发现,这些NAC转录因子基因的启动子区含有多种参与逆境胁迫及激素响应的顺式作用元件,其中TaNAC048-A1基因所含此类调控元件数量最为丰富.低温胁迫下,蚂蚱麦中TaN-AC048-D1、TaNAC048-A1、TaNAC015-B4、TaNAC039-D1、TaNAC104-D2、TaNAC048-B1、TaNAC001-B1 和TaNAC001-A1均显著上调表达;济麦22中,TaNAC048-D1、TaNAC039-D1和TaNAC104-D2基因表达显著上调,TaNAC048-B1基因表达显著下调,其余4个基因的表达无显著变化.采用qRT-PCR验证转录组测序数据,这些TaNAC转录因子基因可能在小麦低温胁迫响应中发挥重要作用.
To investigate the functional characteristics of low-temperature tolerance-related NAC tran-scription factors in wheat,a low-temperature-tolerant variety Mazhamai and a low-temperature-sensi-tive variety Jimai 22 were used for transcriptome sequencing,and 31 candidate NAC transcription fac-tors potentially associated with low-temperature tolerance were screened.Bioinformatics analysis showed that 12 of the TaNAC proteins are basic proteins(pI>7),while the others are acidic proteins(pI<7).All proteins are hydrophilic and mainly localized in the nucleus.The secondary structure was dominated by random coils.Collinearity analysis revealed that there were extensive collinear rela-tionships between NAC transcription factors in wheat and rice.Promoter analysis indicated that the promoter regions of these NAC transcription factor genes contained various cis-acting elements involved in stress responses and hormone signaling,among which TaNAC048-A1 harbored the most abundant regulatory elements.Under low-temperature stress,the expression levels of TaNAC048-D1,TaN-AC048-A1,TaNAC015-B4,TaNAC039-D1,TaNAC104-D2,TaNAC048-B1,TaNAC001-B1,and TaN-AC001-A1 were significantly up-regulated in Mazhamai.In Jimai 22,TaNAC048-D1,TaNAC039-D1,and TaNAC104-D2 were significantly up-regulated,while TaNAC048-B1 was significantly down-regu-lated,and the remaining four genes showed no significant changes.The transcriptome sequencing da-ta were verified by qRTPCR.These TaNAC transcription factor genes may play important roles in the response of wheat to low-temperature stress.
