Modern Research on Astilbin
落新妇苷(astilbin,AST)是土茯苓、黄芪等中药含有的一种二氢黄酮类化合物.近年来研究显示其具有良好的抗氧化、抗炎、免疫调节、调节血糖、神经保护、抗肿瘤、抗高尿酸血症等药理活性,在自身免疫病、代谢性疾病、神经性疾病、癌症等多种疾病的治疗中展现出多途径、多靶点的特点,作用机制涉及Nrf2/HO-1、MAPK/NF-κB/NLRP3、PI3K/AKT、TLR4/MyD88/NF-κB、JAK/STAT、P2Y6R/IL-8/CXCR2等信号通路,具有极大潜力.毒理学研究显示,AST生物安全性良好,无基因毒性、代谢激活及遗传毒性,但药代动力学研究显示,AST存在口服生物利用度差、吸收及消除快、在碱性环境下易被代谢等问题,其代谢产物以3'-O-methylastilbin为主要活性形式,4'-O-methylastilbin为次要活性形式.有研究通过将AST制成纳米制剂,延长半衰期、提高吸收率并改善生物利用度.该文通过检索中国知网、维普、PUBMED、Web of Science等数据库,系统综述AST的药理作用及机制、毒理学、药代动力学及纳米制剂等方面的研究进展,以期为AST药理作用的进一步深入研究、临床应用及新药开发提供借鉴与思路.
Astilbin(AST),a dihydroflavonoid present in traditional Chinese medicinal such as Tufuling(Smilax Glabra)and Huangqi(Astragali Radix).Recent studies have demonstrated that AST exhibits a broad spectrum of favorable pharmacological properties,including antioxidant,anti-inflammatory,immunomodulatory,glucose-regulating,neuroprotective,antitumor,and anti-hyperuricemic activities.It exhibits multi-pathway and multi-target characteristics in the treatment of various diseases,such as autoimmune,metabolic,and neurological disorders,as well as in cancer.These involve a range of signaling pathways,such as Nrf2/HO-1,MAPK/NF-κB/NLRP3,PI3K/AKT,TLR4/MyD88/NF-κB,JAK/STAT,P2Y6R/IL-8/CXCR2.Toxicological assessments indicate that AST possesses a favorable safety profile.However,pharmacokinetic studies highlight several challenges,including low oral bioavailability,rapid absorption and elimination,and instability under alkaline conditions.Among its metabolites,3'-O-methylastilbin serves as the primary active form,with 4'-O-methylastilbin as a secondary active derivative.To overcome these limitations,nanoformulations of AST have been developed,which prolong its half-life,improve absorption,and enhance bioavailability.By comprehensively reviewing literature from databases including CNKI,VIP,PubMed,and Web of Science,this article summarizes recent advances in the pharmacological activities,mechanisms,toxicology,pharmacokinetics,and nanoformulation strategies of AST.We anticipate that this systematic overview will facilitate further exploration of AST's therapeutic potential and support its clinical translation and drug development.
