六种方法构建大鼠盆腔炎模型的比较研究OA

目的 通过比较6种大鼠盆腔炎模型的子宫组织病理形态学变化、炎症因子与粘连相关分子水平,为盆腔炎性疾病及盆腔炎性疾病后遗症动物模型的选择提供参考依据.方法 随机选取10只大鼠设为正常组,余下60只造模大鼠分为6组后分别采用异物置入刺激法、苯酚胶浆致炎法、盐酸致炎法、内源性病原体法、内外源性病原体法、机械损伤加细菌感染法6种不同方法构建大鼠盆腔炎模型,观察子宫组织病理学变化,检测血清中白细胞介素-1α(interleukin-1α,IL-1α)、白细胞介素-6(interleukin-6,IL-6)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、转化生长因子-β(transforming growth factor-β,TGF-β)含量及细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)蛋白表达情况.结果 机械损伤加细菌感染组、内外源性病原体组与盐酸致炎组IL-1α含量均不同程度升高,其余各组IL-1α含量变化不明显;除盐酸致炎组外,其余5组IL-6含量均不同程度升高;除内外源性病原体组外,其余5组VEGF含量均不同程度升高;除盐酸致炎组与内外源性病原体组外,其余4组TGF-β含量均可见不同程度的升高;ICAM-1在各模型组中均呈强阳性表达.病理结果显示,异物置入刺激组及苯酚胶浆致炎组均可见大量结缔组织增生,内膜充血水肿,上皮细胞变形坏死,炎症细胞浸润;盐酸致炎组可见肌层内大量异常血流信号,部分细胞变形坏死;内源性病原体组与内外源病原体组均以炎症细胞增多为主;机械损伤加细菌感染组可见内膜明显充血水肿,上皮细胞部分变形坏死.结论 6种造模方法均可导致大鼠盆腔炎性改变,其中异物置入刺激组、苯酚胶浆致炎组及盐酸致炎组均呈现不同程度的盆腔炎性疾病后遗症表现,以苯酚胶浆致炎组最为典型;内源性病原体组及内外源性病原体组均呈现不同程度的急性盆腔炎表现,以内外源性病原体组最为典型;机械损伤加细菌感染组建立的盆腔炎模型更贴合盆腔炎性疾病后遗症急性发作期表现,适合应用于进一步探索盆腔炎性疾病、盆腔炎性疾病后遗症之间的转变与防治.

Objective To compare the pathological morphological changes of rat uterus tissue,the levels of inflammatory factors and adhesion-related molecules in six rat pelvic inflammatory models,and to provide a reference for the selection of animal models of pelvic inflammatory disease and sequelae of pelvic inflammatory disease.Methods 10 rats were randomly selected as the normal group,while the remaining 60 modeling rats were divided into 6 groups,and pelvic inflammatory models were established by six different methods respectively,including foreign body stimulation,phenol glue inflammation,hydrochloric acid inflammation,endogenous pathogens,exogenous and endogenous pathogens,and mechanical injury plus bacterial infection.The pathological changes of uterine tissue were abserved.The levels of IL-1α,IL-6,VEGF,and TGF-β in serum,and the expression of ICAM-1 protein were obtected.Results The IL-1α content in the mechanical injury combined with bacterial infection group,the exogenous and endogenous pathogen co-infection group and the hydrochloric acid-induced inflammation group increased to varying degrees,while the IL-1α content in the other groups did not change significantly;except for the hydrochloric acid-induced inflammation group,the IL-6 content in the other five groups increased to varying degrees;except for the exogenous and endogenous pathogen co-infection group,the VEGF content in the other five groups increased to varying degrees;except for the hydrochloric acid-induced inflammation group and the exogenous and endogenous pathogen co-infection group,the TGF-β content in the other four groups increased to varying degrees;ICAM-1 was strongly positive in all model groups.Pathological results showed,a large amount of connective tissue hyperplasia,endometrial congestion and edema,epithelial cell deformation and necrosis,and inflammatory cell infiltration were observed in the foreign body implantation stimulation group and the phenol mucilage-induced inflammation group;a large amount of abnormal blood flow signals in the muscular layer were observed in the hydrochloric acid-induced inflammation group,and some cells were deformed and necrotic;the endogenous pathogen infection group and the exogenous and endogenous pathogen co-infection group were mainly characterized by increased inflammatory cells.The mechanical injury combined with bacterial infection group showed obvious endometrial congestion and edema,and some epithelial cells were deformed and necrotic.Conclusion All six modeling methods can lead to pelvic inflammatory changes in rats.Among them,the foreign body implantation stimulation group,phenol mucilage-induced inflammation group and hydrochloric acid-induced inflammation group all showed different degrees of sequelae of pelvic inflammatory disease,and the phenol mucilage-induced inflammation group was the most typical;the endogenous pathogen infection group and the exogenous and endogenous pathogen co-infection group all showed different degrees of acute pelvic inflammatory disease,and the exogenous and endogenous pathogen co-infection group was the most typical;the pelvic inflammatory model established by the mechanical injury combined with bacterial infection group was more consistent with the acute attack of sequelae of pelvic inflammatory disease,and was suitable for further exploration of the transformation and prevention between pelvic inflammatory disease and sequelae of pelvic inflammatory disease.
