鼓室内注射川芎嗪改善顺铂诱导耳毒性大鼠模型作用机制研究OA
Research of Mechanism of Action of Intratympanic Injection of Tetramethylpyrazine in Improving Cisplatin Induced Ototoxicity in Rats
目的 探讨鼓室内注射川芎嗪(tetramethylpyrazine,TMP)干预顺铂诱导耳毒性大鼠模型的作用机制.方法 实验大鼠分为空白组对照组(B组)、空白+鼓室注射TMP组(B+L组)、模型对照组(M组)、模型+鼓室注射TMP组(M+L组).听性脑干反应检测大鼠听力变化;耳蜗基底膜铺片检测大鼠耳蜗毛细胞的病理改变;比色法检测大鼠血清中一氧化氮(nitric oxide,NO)、过氧化氢(hydrogen peroxide,H2O2)和丙二醛(malondialdehyde,MDA)含量及超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性;Western blot法检测大鼠耳蜗组织中细胞质核因子E2相关因子2(nuclear factor erythroid 2-related factor 2,Nrf2)、细胞核Nrf 2、血红素加氧酶-1(heme oxygenase-1,HO-1)、γ-谷氨酰半胱氨酸合成酶(γ-glutamylcysteine synthetase,γ-GCS)蛋白表达;RT-qPCR法检测大鼠耳蜗组织中Nrf 2、HO-1和γ-GCS mRNA水平.结果 与B组比较,M组大鼠听力阈值升高,毛细胞缺失,血清中NO含量降低,H2O2、MDA含量升高,抗氧化酶活性降低,耳蜗组织细胞质中Nrf 2蛋白表达增高,细胞核中Nrf 2、HO-1、γ-GCS蛋白表达显著降低(P<0.05);与M组比较,M+L组大鼠听力阈值降低,毛细胞损失减轻,血清中NO含量增高,H2O2、MDA含量降低,抗氧化酶活性增高,耳蜗组织细胞质中Nrf 2蛋白表达降低,细胞核中Nrf 2、HO-1、γ-GCS蛋白表达显著升高(P<0.05).结论 鼓室内注射TMP可以通过调控Nrf 2/抗氧化反应元件(antioxidant response element,ARE)信号通路,促进Nrf 2核转移,增加HO-1、γ-GCS蛋白的表达,从而减轻顺铂诱导的耳毒性.
Objective To explore the mechanism of intratympanic injection of tetramethylpyrazine(TMP)in intervention of cisplatin induced ototoxicity in rats.Methods The experimental rats were divided into blank control group(group B),blank+tympanic injection TMP group(group B+L),model control group(group M),and model+tympanic injection TMP group(group M+L).Auditory brainstem response was used to detect hearing changes in rats;Using cochlear basement membrane preparation to detect the pathological changes in rat cochlear hair cells;The colorimetric method was used to detect the levels of nitric oxide(NO),hydrogen peroxide(H2O2),and malondialdehyde(MDA)in rat serum,as well as the activities of superoxide dismutase(SOD),catalase(CAT),and glutathione peroxidase(GSH-Px);Western blot was used to detect the expression of cytoplasmic nuclear factor erythroid 2-related factor 2(Nrf2),nuclear Nrf2,heme oxygenase-1(HO-1),gamma glutamylcysteine synthase(γ-GCS)protein in rat cochlear tissue;RT-qPCR was used to detect the mRNA levels of Nrf2,HO-1,and γ-GCS in rat cochlear tissue.Results Compared with group B,the hearing threshold of rats in group M increased,hair cells were lost,the content of NO in serum decreased,the contents of H2O2 and MDA increased,the activity of antioxidant enzymes decreased,the expression of Nrf2 protein in the cytoplasm of cochlear tissue increased,and the expressions of Nrf2,HO-1 and γ-GCS proteins in the nucleus significantly decreased(P<0.05).Compared with group M,the hearing threshold of rats in group M+L decreased,the loss of hair cells was alleviated,the content of NO in serum increased,the contents of H2O2 and MDA decreased,the activity of antioxidant enzymes increased,the expression of Nrf2 protein in the cytoplasm of cochlear tissue decreased,and the expressions of Nrf2,HO-1 and γ-GCS proteins in the nucleus significantly increased(P<0.05).Conclusion Intratympanic injection of TMP can promote Nrf2 nuclear translocation,increase the expression of HO-1 and γ-GCS proteins,and alleviate cisplatin induced ototoxicity by regulating the Nrf2/antioxidant response element(ARE)signaling pathway.
