miR-892b对骨肉瘤细胞增殖、迁移和凋亡的影响及其机制OA
Effect and mechanism of miR-892b on the proliferation,migration,and apoptosis of osteosarcoma cells
目的 探究miR-892b对骨肉瘤细胞增殖、迁移和凋亡的影响及其机制.方法 通过RT-qPCR实验检测人骨肉瘤细胞系(MG-63、U2OS)以及人成骨细胞系(hFOB1.19)中miR-892b的相对表达水平.将U2OS细胞分为A~D组,分别转染阴性对照模拟物(A组)、miR-892b模拟物(B组)、阴性对照抑制物(C组)和miR-892b抑制物(D组).通过CCK-8实验检测转染后培养第24、48、72小时时各组的吸光度值.通过Transwell实验检测各组的迁移细胞数.通过Western blot(WB)实验检测各组细胞中凋亡相关蛋白Bax和NF-κB信号通路中关键蛋白p-NF-κB的表达水平.结果 RT-qPCR实验结果显示,与hFOB1.19细胞相比,MG-63细胞和U2OS细胞中miR-892b的相对表达水平显著降低(F=73.59,tDunnett=7.83、9.92,P<0.05).CCK-8实验结果显示,培养至第24、48、72小时时,B组吸光度值显著低于A组,而D组吸光度值显著高于 C组(F=5.91~31.23,P<0.05).与 A组相比,B组迁移细胞数显著减少,Bax表达水平显著升高,p-NF-κB表达水平显著降低;与 C组相比,D组迁移细胞数显著增加,Bax表达水平显著降低,p-NF-κB表达水平显著升高(t=3.64~7.16,P<0.05).结论 miR-892在骨肉瘤细胞中呈低表达,且可能通过影响NF-κB通路来调控骨肉瘤细胞的增殖、迁移和凋亡.
Objective To investigate the effect and mechanism of miR-892b on the proliferation,migration,and apopto-sis of osteosarcoma cells.Methods RT-qPCR was used to measure the relative expression level of miR-892b in osteosarcoma cell lines(MG-63,U2OS)and the human osteoblast cell line hFOB1.19.U2OS cells were divided into group A(transfected with mimic NC),group B(transfected with miR-892b mimic),group C(transfected with inhibitor NC),and group D(transfected with miR-892b inhibitor).CCK-8 assay was used to measure the absorbance value of each group at 24,48,and 72 hours of culture after transfection;Transwell assay was used to measure the number of migrating cells in each group;Western blot was used to measure the expression levels of the apoptosis-related protein Bax and the key protein p-NF-κB in the NF-κB signaling pathway.Results RT-qPCR showed that compared with the hFOB1.19 cells,the MG-63 and U2OS cells showed a significant reduction in the relative expression level of miR-892b(F=73.59,tDunnett=7.83,9.92,P<0.05).CCK-8 assay showed that at 24,48,and 72 hours of cul-ture,group B had a significantly lower absorbance value than group A,while group D had a significantly higher absorbance value than group C(F=5.91-31.23,P<0.05).Compared with group A,group B had a significant reduction in the number of migrating cells,a significant increase in the protein expression level of Bax,and a significant reduction in the protein expression level of p-NF-κB;compared with group C,group D had a significant increase in the number of migrating cells,a significant reduction in the pro-tein expression level of Bax,and a significant increase in the protein expression level of p-NF-κB(t=3.64-7.16,P<0.05).Con-clusion There is a relatively low expression level of miR-892b in osteosarcoma cells,and miR-892b may regulate the proliferation,migration,and apoptosis of osteosarcoma cells by affecting the NF-κB pathway.
