基于辛酸-乙酸协同除杂的抗毒素血清制备工艺优化及其关键质量指标分析OA
Optimization of antitoxin serum preparation process based on caprylic acid-acetic acid synergistic impurity removal and analysis of its key quality indicators
基于辛酸-乙酸协同除杂机制,建立了一种操作简便、原料易得、生产成本低廉的抗毒素血清制备方法.实验中分别制备得到了抗破伤风血清、抗狂犬病血清和抗蝮蛇毒血清,与同等条件下的传统制备工艺相比,抗破伤风血清的 F(ab')2 含量提升 19%~27%(温度稳定性提高 3.6%~7%)、比活性提升 55.8%~94.7%(温度稳定性明显提升 17.5%)、效价提升 12.9%~40.0%(温度稳定性更加显著地提升52%).该方法在抗狂犬病血清及抗蝮蛇毒血清的制备中同样表现出优越性(F(ab')2 含量>85%、稳定性>84.7%).研究结果表明,该方法适用于多种抗毒素血清的制备,所得产品具有安全性能、F(ab')2 含量和比活性都高且稳定性好的特点.通过对关键工艺参数的优化及关键质量指标的全面分析,验证了该方法的普适性与可靠性.本研究成果为推动抗毒素血清纯化技术的进一步提升提供了理论依据和实践参考.
In this study,a simple operation,easily accessible,and low-cost antitoxin serum preparation method was established based on the caprylic acid-acetic acid synergistic impurity removal mechanism.Experiments were performed to prepare tetanus antitoxin serum,rabies antitoxin serum and Agkistrodon halys antivenin,respectively.Under identical experimental conditions,comparison with the traditional preparation process revealed that tetanus antitoxin serum exhibited a 19%-27%increase in F(ab')₂ content(with temperature stability improved by 3.6%-7%),a 55.8%-94.7%increase in specific activity(with temperature stability enhanced by up to 17.5%),and a 12.9%-40.0%increase in potency(with temperature stability elevated by up to 52%).Meanwhile,this method also showed superior performance in the preparation of rabies antitoxin serum and A.halys antivenin(F(ab')₂ content>85%,stability>84.7%).The results demonstrated that this method is applicable to the preparation of various antitoxin sera,yielding products with high safety,elevated F(ab')₂content,enhanced specific activity,and favorable stability.Through optimizing the critical process parameters and comprehensively analyzing the core quality indicators,the universality and reliability of the established method were validated.This study provides a theoretical basis and practical reference for further opimizing the purification technology of antitoxin serum.
