目的 探讨龟板固本方对环磷酰胺(CTX)诱导的免疫抑制小鼠的免疫调节作用.方法 将C57BL/6J小鼠按体质量随机分为正常对照组、模型组、胸腺肽组[100 μg/(kg·d)]、龟板固本方低剂量组[5 g/(kg·d)]、龟板固本方高剂量组[20 g/(kg·d)],每组10只.胸腺肽组腹腔注射相应剂量的胸腺肽,龟板固本方低、高剂量组灌胃相应剂量的龟板固本方,正常组和模型组灌胃等量生理盐水,1次/d,连续7d.除正常对照组外,其余各组小鼠在最后3d内连续腹腔注射CTX,剂量为80 mg/(kg·d),以建立免疫抑制小鼠模型.测定各组小鼠的体质量及脾脏指数、胸腺指数、肝脏指数;全自动血液分析仪检测各组小鼠白细胞数量、红细胞数量及血小板数量;瑞氏染色检测各组小鼠巨噬细胞吞噬率和巨噬细胞吞噬指数;MTT检测各组小鼠T淋巴细胞刺激指数;乳酸脱氢酶(LDH)测定各组小鼠自然杀伤(NK)细胞杀伤活性;ELISA测定各组小鼠血清免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、白细胞介素(IL)-3、IL-1β、IL-4、IL-6和γ干扰素(IFN-γ)水平.结果 与正常对照组比较,模型组小鼠体质量、脾脏指数、胸腺指数、肝脏指数、白细胞数量、红细胞数量、血小板数量、巨噬细胞吞噬率和巨噬细胞吞噬指数、T淋巴细胞刺激指数、NK细胞杀伤活性及血清IgG、IgM、IL-3、IL-1β、IL-4、IL-6、IFN-γ水平均显著降低(P<0.01);与模型组比较,胸腺肽组和龟板固本方高剂量组小鼠体质量、脾脏指数、胸腺指数、肝脏指数、白细胞数量、红细胞数量、血小板数量、巨噬细胞吞噬率和巨噬细胞吞噬指数、T淋巴细胞刺激指数、NK细胞杀伤活性及血清IgG、IgM、IL-3、IL-1β、IL-4、IL-6、IFN-γ水平均显著升高(P<0.05,P<0.01);与胸腺肽组比较,龟板固本方低剂量组小鼠体质量、白细胞数量、血小板数量、巨噬细胞吞噬率及巨噬细胞吞噬指数、T淋巴细胞刺激指数、NK细胞杀伤活性及血清IgM水平均显著降低(P<0.05,P<0.01);与龟板固本方低剂量组比较,龟板固本方高剂量组小鼠体质量、脾脏指数、胸腺指数、肝脏指数、白细胞数量、血小板数量、巨噬细胞吞噬率及巨噬细胞吞噬指数、T淋巴细胞刺激指数、NK细胞杀伤活性和血清IgG、IgM水平均显著升高(P<0.05,P<0.01).结论 龟板固本方通过改善免疫系统相关指标,有效增强了CTX诱导的免疫抑制小鼠的免疫功能.

Objective To explore the immunomodulatory effect of Guiban Guben Formula(GBGBF)on cyclophosphamide(CTX)-induced immunosuppressed mice.Methods C57BL/6J mice were randomly divided by body weight into normal control group,model group,thymosin group(0.2 mL/d),low-dose GBGBF group(0.5 mL/d),and high-dose GBGBF group(1.0 mL/d),with 10 mice per group.The thymosin group received intraperitoneal injections of the corresponding dose of thymosin;the low-and high-dose GBGBF groups were administered the corresponding doses of the GBGBF via gavage;and the normal control and model groups were administered the same volume of saline via gavage,once daily for 7 consecutive days.Except for the normal control group,mice in the other groups were intraperitoneally injected with CTX at a dose of 80 mg/kg·d-1 for the last 3 days to establish an immunosuppressed mouse model.Body weight,spleen index,thymus index,and liver index were measured for each group.An automatic hematology analyzer was used to determine the counts of white blood cells,red blood cells,and platelets.Wright's staining was performed to measure macrophage phagocytic rate and phagocytic index.The MTT assay was used to evaluate T lymphocyte stimulation index.Lactate dehydrogenase(LDH)assay was conducted to check natural killer(NK)cell cytotoxic activity.ELISA was employed to determine serum levels of immunoglobulin G(IgG),immunoglobulin M(IgM),interleukin(IL)-3,IL-1β,IL-4,IL-6,and interferon-γ(IFN-γ)in each group.Results Compared with the normal control group,the model group exhibited significant decreases in body weight,spleen index,thymus index,liver index,white blood cell count,red blood cell count,platelet count,macrophage phagocytic rate,macrophage phagocytic index,T lymphocyte stimulation index,and NK cell cytotoxic activity,as well as serum levels of IgG,IgM,IL-3,IL-1β,IL-4,IL-6,and IFN-γ(P<0.01).Compared with the model group,both the thymosin group and the high-dose GBGBF groups showed significant increases in body weight,spleen index,thymus index,and liver index,white blood cell count,red blood cell count,platelet count,macrophage phagocytic rate,macrophage phagocytic index,T lymphocyte stimulation index,and NK cell cytotoxic activity,as well as serum levels of IgG,IgM,IL-3,IL-1β,IL-4,IL-6,and IFN-γ(P<0.05,P<0.01).Compared with the thymosin group,the low-dose GBGBF group showed significantly lower body weight,white blood cell count,platelet count,macrophage phagocytic rate,macrophage phagocytic index,T lymphocyte stimulation index,NK cell cytotoxic activity,and serum IgM levels(P<0.05,P<0.01).Compared with the low-dose GBGBF group,the high-dose GBGBF group demonstrated significant increases in body weight,spleen index,thymus index,liver index,white blood cell count,platelet count,macrophage phagocytic rate,macrophage phagocytic index,T lymphocyte stimulation index,NK cell cytotoxic activity,and serum levels IgG and IgM(P<0.05,P<0.01).Conclusion GBGBF effectively enhances the immune function of CTX-induced immun-osuppressed mice by improving immune system-related indicators.
