孕酮微胶囊冻干工艺的优化及其体外释放研究OA
Study on the optimization of lyophilization process and in vitro release of progesterone microcapsules
为优化孕酮微胶囊的真空冷冻干燥工艺并评价其体外释放特性,试验系统考察了关键冻干参数对孕酮微胶囊的保护效果,设置了预冻时间梯度(2,4,6,8,12 h)和预冻温度梯度(-20,80℃),并筛选了不同浓度(浓度分别为3.0%、5.0%、7.5%)的蔗糖、葡萄糖和乳糖作为冻干保护剂,以筛选最佳预冻条件与冻干保护剂,并对孕酮微胶囊的外观形态、再分散性、复溶时间、包封率及载药量等核心质量指标进行全面评价;同时采用模拟反刍动物三阶段体外消化模型(瘤胃液-胃液-小肠液)评估其稳定性与靶向释放性.结果表明:在预冻时间为 8 h、预冻温度为-80℃条件下,孕酮微胶囊外观平整饱满、气泡少且无喷溅现象;添加3.0%乳糖作为保护剂时,孕酮微胶囊结构疏松多孔,包封率为96.68%,载药量为 19.97%,显著高于空白对照(P<0.05).孕酮微胶囊在模拟瘤胃液和胃液中 24 h 内累积释放率仅分别为 28.74%和 29.15%,而在模拟小肠液中 10 h 内累积释放率为 80.61%,显示出良好的瘤胃-胃保护与小肠靶向释放特性.说明本研究确立的优化工艺(预冻温度为-80℃、预冻时间为 8 h、冻干保护剂为 3.0%乳糖)能制备出性能优良的冻干孕酮微胶囊,为反刍动物同期发情口服制剂的开发提供重要依据.
In order to optimize the vacuum freeze-drying process of progesterone microcapsules and evaluate their in vitro release characteristics,the protective effects of key freeze-drying parameters on progesterone microcapsules were systematically investigated.The pre-freezing time gradient(2,4,6,8 and 12 h)and pre-freezing temperature gradient(-20 and 80℃)were set up,and sucrose,glucose and lactose with different concentrations(3.0%,5.0%and 7.5%,respectively)were selected as freeze-drying protective agents to screen the best pre-freezing conditions and freeze-drying protective agents.The core quality indicators such as appearance,redispersibility,reconstitution time encapsulation efficiency and drug loading of progesterone microcapsules were comprehensively evaluated.At the same time,the three-stage in vitro digestion model(rumen fluid-gastric fluid-small intestinal fluid)of simulated ruminants was used to evaluate its stability and targeted release.The results showed that under the conditions of pre-freezing time of 8 h and pre-freezing temperature of-80℃,the progesterone microcapsules had a smooth and full appearance,few bubbles and no splashing.When 3.0%lactose was added as a protective agent,the structure of progesterone microcapsules was loose and porous;the encapsulation rate was 96.68%,and the drug loading was 19.97%,which was significantly higher than that of the blank control(P<0.05).The cumulative release rates of progesterone microcapsules in simulated rumen fluid and gastric fluid within 24 h were only 28.74%and 29.15%,respectively,while the cumulative release rate in simulated intestinal fluid within 10 h was 80.61%,showing good rumen-stomach protection and small intestine targeted release characteristics.The results showed that the optimized process(pre-freezing temperature was-80℃,pre-freezing time was 8 h,and freeze-dried protective agent was 3.0%lactose)established in this study could prepare freeze-dried progesterone microcapsules with excellent performance,which provided an important basis for the development of estrus synchronization oral preparations for ruminants.
